SIRT1调节线粒体自噬对猪肠上皮细胞氧化损伤的影响 SIRT1(Sirtuin1)isakeyproteininvolvedintheregulationofcellularprocessessuchasmetabolism,cellsurvival,andstressresponse.Ithasbeenextensivelystudiedinvariousbiologicalsystems,includingtheregulationofmitochondrialfunctionandautophagy.Inrecentyears,researchershavefocusedonunderstandingtheroleofSIRT1inoxidativedamageanditseffectsonintestinalepithelialcellsinpigs. Oxidativedamageinintestinalepithelialcellscanoccurduetovariousfactors,includingexposuretoreactiveoxygenspecies(ROS)andinflammation.Theintegrityandfunctionoftheintestinalepithelialbarrierarecrucialformaintainingguthomeostasisandpreventingtheentryofharmfulsubstances.Oxidativedamagetotheintestinalepitheliumcanimpairbarrierfunctionandleadtothedevelopmentofvariousdiseases,includinginflammatoryboweldisease(IBD). Autophagyisahighlyregulatedprocessresponsibleforthedegradationandrecyclingofdamagedorunnecessarycellularcomponents.Itplaysacrucialroleinmaintainingcellularhomeostasisandpromotingcellsurvivalunderstressfulconditionssuchasoxidativedamage.RecentstudieshaveshownthatSIRT1playsapivotalroleintheregulationofautophagyinvariouscelltypes,includingintestinalepithelialcells. SIRT1actsasapositiveregulatorofautophagybydirectlydeacetylatingandactivatingkeyautophagy-relatedproteins,suchasAtg5andAtg7.ActivationofautophagythroughSIRT1canhelpremovedamagedmitochondriaandpreventexcessiveROSproduction,therebyreducingoxidativedamageinintestinalepithelialcells.Additionally,SIRT1hasbeenshowntoinhibitinflammationandpromotecellsurvivalunderoxidativestressconditions. SeveralstudieshavedemonstratedtheprotectiveeffectsofSIRT1-mediatedautophagyinthepreventionofoxidativedamageinintestinalepithelialcells.Forexample,onestudyusingaporcinemodelshowedthatSIRT1activationattenuatedoxidativedamageinducedbyhydrogenperoxide(H2O2)inintestinalepithelialcells.ActivationofSIRT1ledtoincreasedautophagy,whichenhancedtheclearanceofdamagedmitochondriaandreducedROSaccumulation. Furthermore,anotherstudyfoundthatSIRT1activationprotectedag
