东方白鹳微卫星遗传标记的筛选及其在种群遗传分析中的应用(英文) Introduction TheOrientalWhiteStork(Ciconiaboyciana)isabirdspeciesnativetoEastAsia,withapopulationthathasexperiencedadeclineoverthepastfewdecades.ThespeciesislistedasEndangeredontheInternationalUnionforConservationofNature(IUCN)RedListduetohabitatloss,hunting,andpollution.Todevelopeffectiveconservationstrategies,itisessentialtogainabetterunderstandingofthegeneticstructureanddiversityofthespecies.Microsatellitemarkersareusefultoolsforstudyingpopulationgenetics,butthedevelopmentofanewsetofmarkerswouldhelpprovidehigherresolutionanalysisofthespecies.Inthisstudy,wedevelopedasetofmicrosatellitemarkersusingnext-generationsequencingtechnology,whichwerescreenedandevaluatedfortheirsuitabilityforgeneticanalysisoftheOrientalWhiteStorkpopulations. Methods Bloodsampleswerecollectedfrom30OrientalWhiteStorkindividualsfromtwopopulations:NanchangandTaiheinJiangxiprovince,China.DNAextractionwasperformedusingtheQIAampDNABloodMiniKit.Microsatelliteswereidentifiedfromnext-generationsequencingdata,andatotalof100microsatelliteswereselectedforprimerdesign.Polymerasechainreaction(PCR)amplificationwasperformedusingthefluorescent-labeledprimers,andgenotypingwasconductedusinganABIPRISM3730DNAanalyzer.Populationgeneticanalysiswasperformedusingvariousstatisticalmethods,includinggeneticdiversity,populationstructure,andgeneflowanalysis. Results Outofthe100microsatellitemarkersinitiallyselected,20markerswithhighpolymorphismandclearamplificationwerechosenforfurtheranalysis.TheresultsshowedthattheOrientalWhiteStorkpopulationshadmoderatelevelsofgeneticdiversity(meanexpectedheterozygosityof0.694),withnosignificantdifferencesobservedbetweenthetwopopulations.Inthepopulationstructureanalysis,theresultsindicatedthatthetwopopulationswereseparatebutshowedsomeadmixture.Additionally,geneflowanalysissuggestedthattheremightbeacertainlevelofgeneflowbetweenthetwopopulations. Discussion Inthisstudy,wedevelopedasetofnewmicrosatellitemarkersandevaluatedtheirsuitabilityforgeneticanalysiso
