Objectives: 1.ToconstructamicrosatelliteenrichmentlibraryfromOFMgenomicDNA. 2.ToscreenthemicrosatellitemarkersandselectthemostinformativeonesforfurtheruseingeneticstudiesofOFM. Methods: 1.GenomicDNAextraction:GenomicDNAwillbeisolatedfromadultOFMindividualsusingastandardprotocol. 2.Constructionofmicrosatelliteenrichmentlibrary:Thelibrarywillbeconstructedusingaprotocolthatinvolvestheenrichmentofmicrosatelliterepeatsusingbiotin-labeledprobes,followedbyPCRamplificationandcloningoftheenrichedDNAfragments. 3.Microsatellitemarkerscreening:ThelibrarywillbescreenedusingPCRamplificationandsequencingoftheenrichedDNAfragments.TheresultingsequenceswillbeanalyzedforthepresenceofSSRsusingappropriatesoftware.PrimerpairswillbedesignedforamplifyingtheselectedSSRmarkers,andtheirpolymorphismandinformativenesswillbeevaluatedusingapanelofOFMindividuals. Expectedoutcomes: 1.AmicrosatelliteenrichmentlibraryofOFMwillbeconstructedandevaluatedforitsefficiencyincapturingSSRs. 2.AsetofinformativeandpolymorphicSSRmarkerswillbeidentifiedandvalidatedforuseingeneticstudiesofOFM.
