绍兴鸭热休克基因研究以及对细胞凋亡相关基因表达的影响 Introduction Heatshockproteins(HSPs)areagroupofmolecularchaperonesthathelptopreventdamagetocellsunderstressconditions.Inanimals,severalHSPgeneshavebeencharacterized,includingHSP90,HSP70,andHSP60.Thesegenesareinvolvedinvariousphysiologicalprocessessuchasproteinsynthesis,folding,translocation,anddegradation.TheexpressionofHSPsisregulatedbyvarioussignalingpathways,includingheatshockfactor(HSF)andnuclearfactor-kappaB(NF-κB).Inrecentyears,severalstudieshaveinvestigatedtheroleofHSPgenesintheadaptationofanimalstoheatstress.ThisresearchaimstoinvestigatetheexpressionofHSPgenesintheliverofducksunderheatstress,aswellastheireffectsoncellapoptosis-relatedgenes. MaterialsandMethods Inthisstudy,weused20healthyducks(maleandfemale,3monthsold)fromafarminShaoxing,China.Theywereallocatedintotwogroupsrandomly:controlgroup(n=10),whichwasmaintainedatanambienttemperatureof25°C,andheatshockgroup(n=10),whichwassubjectedtoahigh-temperatureenvironment(38°C)for2hours.Afterthetreatment,allducksweresacrificed,andthelivertissueswereimmediatelycollectedforgeneexpressionanalysis.TotalRNAwasextractedusingTrizolreagent,andthequalitywascheckedbyagarosegelelectrophoresis.Quantitativereal-timePCRwasperformedtomeasurethemRNAexpressionlevelsofHSP90,HSP70,HSP60,Bax,Bcl-2,andCaspase-3.Relativegeneexpressionwascalculatedusingthe2^-△△Ctmethod. Results TheresultsshowedthattheexpressionlevelsofHSP90,HSP70,andHSP60weresignificantlyupregulatedintheheatshockgroupcomparedwiththecontrolgroup.Specifically,theHSP90mRNAlevelincreasedby6.2-fold,HSP70levelby4.3-fold,andHSP60levelby2.5-foldintheheatshockgroup.Inaddition,themRNAexpressionlevelsofBaxandCaspase-3weresignificantlyhigherintheheatshockgroupthaninthecontrolgroup,whiletheexpressionlevelofBcl-2wassignificantlydecreased.TheBax/Bcl-2ratio,whichisamarkerforcellapoptosis,wassignificantlyhigherintheheatshockgroupthaninthecontrolgroup.
