Introduction: Ganodermasporepolysaccharidesarehigh-molecular-weightcompoundsknownfortheirimmunomodulatory,antioxidant,andanticanceractivities.However,theextractionofthesepolysaccharidesfromGanodermasporescanbechallengingduetothecomplexstructureandlowsolubility.Thus,optimizationoftheextractionprocessiscrucialforobtainingahighyieldofGanodermasporepolysaccharides. Methods: Responsesurfacemethodology(RSM)isastatisticaltechniqueusedformodelingandoptimizingcomplexprocesses.Inthisstudy,RSMwasemployedtooptimizethehot-waterextractionprocessforGanodermasporepolysaccharides.Theextractionparametersincludingextractiontemperature,extractiontime,andwater-to-sporeratiowereselectedasindependentvariables.TheresponsevariablewastheyieldofGanodermasporepolysaccharides. Results: Basedonacentralcompositedesign(CCD)matrix,atotalof20experimentswereconductedtoevaluatetheeffectsoftheextractionparametersontheyieldofGanodermasporepolysaccharides.Theexperimentalresultswerefittedintoasecond-orderpolynomialmodelusingmultipleregressionanalysis.Analysisofvariance(ANOVA)wasperformedtodeterminethesignificanceofthemodelandtheindividualfactors. Discussion: TheresultsoftheANOVAindicatedthatallthreefactors(extractiontemperature,extractiontime,andwater-to-sporeratio)significantlyinfluencedtheyieldofGanodermasporepolysaccharides(p<0.05).Amongthem,extractiontemperaturehadthemostsignificanteffectontheyield,followedbyextractiontimeandwater-to-sporeratio.Theoptimizedparametersforachievingmaximumyieldwerefo
