Introduction Embryonicstemcells(ESCs)arepluripotentcellsthatcandifferentiateintoanycelltypeinthebody.TheregulationofESCdifferentiationisacomplexprocessthatinvolvestheactivationandrepressionofspecificgeneexpressionprograms.ThemaintenanceofESCpluripotencyinvolvestheinteractionofvarioustranscriptionalfactors,includingOct4,Sox2,andNanog,whichareresponsiblefortheactivationoftargetgenesrequiredformaintainingthepluripotentstate.However,thedifferentiationofESCsrequiresthesuppressionofthesefactorsandtheactivationofanewsetofgenesthatareresponsiblefortheformationofdifferentcelltypes. Severalstudieshaveshownthatthehistoneacetyltransferasep300playsanimportantroleintheregulationofESCdifferentiation.P300isatranscriptionalcoactivatorthatbindstospecifictranscriptionfactorsandpromoteschromatinremodelingthroughitsabilitytoacetylateand/ordeacetylatehistones,resultingintheactivationorrepressionofgeneexpression.Additionally,p300interactswithotherepigeneticmodifiers,suchasDNAmethyltransferasesandhistonemethyltransferases,toregulategeneexpressionandcellfatedecisions.
