羟乙基淀粉对重症急性胰腺炎大鼠肝脏水通道蛋白-1表达及肝脏损伤的影响 Introduction Severeacutepancreatitis(SAP)isalife-threateningdiseasecharacterizedbypancreaticnecrosisandsubsequentsystemicinflammatoryresponsesyndrome(SIRS)leadingtomultipleorgandysfunction.OneoftheorganscommonlyaffectedinSAPistheliver.LiverdamageinSAPischaracterizedbyincreasedserumlevelsofliverenzymesandbilirubin,alongwithhistologicalchangessuchassinusoidalcongestion,hemorrhage,andinflammatorycellinfiltration.HepaticinjuryinSAPisthoughttobecausedbyacombinationoffactors,suchaslocalinflammation,oxidativestress,andsystemicinflammation.Theaquaporin1(AQP1)isawaterchannelexpressedintheliverthatcontributestohepaticwaterhomeostasisandparticipatesinmultiplesignalingpathways.Inthepresentstudy,weinvestigatedtheeffectsofhydroxyethylstarch(HES)onAQP1expressionandliverinjuryinSAPrats. Methods Thirty-twomaleSprague-Dawleyratsweighing250-300gwererandomlydividedintofourgroups:controlgroup(n=8),SAPgroup(n=8),SAP+HESgroup(n=8),andHESgroup(n=8).SAPwasinducedbyretrogradeinjectionof5%sodiumtaurocholateintothebiliopancreaticduct.TheHESgroupreceivedasingleintravenousdoseof10mL/kgHES(6%hydroxyethylstarch130/0.4solution)immediatelyafterSAPinduction.TheSAP+HESgroupreceivedthesamedoseofHESplusstandardsupportivetherapy(SST)consistingoffluidresuscitation,antibiotics,analgesics,andnutritionalsupport.ThecontrolgroupreceivedthesamesurgicalprocedurewithoutSAPinductionorHESadministration.After24hours,theratsweresacrificed,andbloodandlivertissuesampleswerecollectedforfurtheranalysis.Serumlevelsofalaninetransaminase(ALT),aspartatetransaminase(AST),andtotalbilirubin(TBIL)weremeasuredusingcommercialkits.Livertissuesampleswerefixedin10%formalinandprocessedforhematoxylin-eosin(HE)stainingandimmunohistochemistryforAQP1.AQP1expressioninlivertissuewasscoredbasedonthepercentageofpositivecellsandintensityofstaining.
