大黄素通过ILK通路调控上皮性卵巢癌细胞迁移、侵袭及其机制的实验研究 Title:ExperimentalStudyontheRegulationofEpithelialOvarianCancerCellMigrationandInvasionbyILKPathwayandtheMechanismofActionbyDaHuangSu Abstract: Epithelialovariancancer(EOC)isoneoftheleadingcausesofcancer-relateddeathsinwomenworldwide.Despiteadvancesintreatmentstrategies,theprognosisforEOCremainspoorduetoitsaggressivenatureandhightendencyformetastasis.Therefore,thereisanurgentneedtoexplorenoveltherapeutictargetsforEOC.Inthisstudy,weinvestigatedtheroleoftheintegrin-linkedkinase(ILK)pathwayinregulatingEOCcellmigrationandinvasionandexploredthemechanismofactionofDaHuangSu,atraditionalChinesemedicine,intargetingthispathway. Introduction: TheILKpathwayisknowntoplayacrucialroleintheregulationofcelladhesion,migration,andinvasion.AberrantactivationoftheILKpathwayhasbeenimplicatedinvariouscancers,includingEOC.DaHuangSu,atraditionalChinesemedicinederivedfromtherhubarbplant,hasshownpromisinganti-cancereffects.However,itsmechanismofactioninEOCisnotfullyunderstood. Methods: 1.CellCultureandExperimentalDesign:EOCcelllineswereculturedanddividedintodifferenttreatmentgroups,includingcontrol,ILKoverexpression,ILKknockdown,andDaHuangSutreatment. 2.CellMigrationandInvasionAssays:TranswellmigrationandinvasionassayswereperformedtoassesstheeffectofILKpathwaymodulationandDaHuangSutreatmentonEOCcellmigrationandinvasion. 3.WesternBlotAnalysis:ProteinexpressionlevelsofILKpathwaycomponents,EMTmarkers,andmatrixmetalloproteinases(MMPs)wereanalyzedbyWesternblottingtoinvestigatetheunderlyingmechanismofaction. Results: 1.ILKOverexpressionPromotedEOCCellMigrationandInvasion:OverexpressionofILKsignificantlyincreasedthemigratoryandinvasivecapabilitiesofEOCcellscomparedtothecontrolgroup. 2.ILKKnockdownInhibitedEOCCellMigrationandInvasion:KnockdownofILKattenuatedEOCcellmigrationandinvasioncomparedtothecontrolgroup. 3.DaHuangSuSuppressedEOCCellMigrationandInvasionviaILKPathway:TreatmentwithDaHuangSusignificantlyinhibitedthemigratoryandinvasivecapabilitiesofEOCcells.Furthe
