TheHenryreactioninvolvestheadditionofanucleophiletoanaldehydeoraketonetoformanewcarbon-carbonbond.Itisanimportantreactioninorganicsynthesis,asitallowstheconstructionofchiralβ-hydroxyketonesandalcohols,whichareversatilebuildingblocksforthesynthesisofpharmaceuticals,naturalproducts,andotherorganicmolecules. OneofthechallengesintheHenryreactioniscontrollingthestereochemistryoftheproduct.Traditionalapproachesinvolveusingchiralauxiliariesorchiralcatalyststoinduceasymmetry.However,chiralnitrogen-containingalcoholshaveemergedasapromisingalternativeduetotheirabilitytochelatetometalcatalystsandcontrolthestereochemistryofthereaction. Zinc-catalyzedasymmetricHenryreactionsusingchiralnitrogen-containingalcoholsasligandshavebeenreportedinanumberofstudies.Forexample,Liuandco-workersreportedthesynthesisofβ-hydroxyketoneswithhighenantioselectivityusingachiralnitrogen-containingalcoholligandinthepresenceofazinccatalyst.Theauthorsfoundthatthereactionproceededefficientlyundermildconditions,andtheresultingproductswereobtainedinhighyieldandwithgoodstereocontrol. Similarly,Shiandco-workersreportedthesynthesisofchiralβ-hydroxyindanoneswithhighenantioselectivityusingachiralnitrogen-containingalcoholligandinthepresenceofazinccatalyst.Theauthorsfoundthatthepresenceofthechiralligandwasessentialforcontrollingthestereochemistryofthereactionandachievinghighenantioselectivity.
