基于高通量测序技术研究水貂远端肠道细菌群落组成(英文) Introduction Mustelavison,commonlyknownastheAmericanmink,isasemi-aquaticmammalnativetoNorthAmerica.ItbelongstothefamilyMustelidae,whichalsoincludesotherspeciessuchasotters,weasels,andferrets.Theminkisanimportantfur-bearinganimalthathasbeenextensivelyfarmedforitsfur,butitisalsoapotentialreservoirforzoonoticpathogens.Therefore,itisimportanttounderstanditsgutmicrobiotatomaintainitshealthandreducetheriskoftransmittingpotentiallyharmfulbacteria.Inthisstudy,weaimedtoinvestigatethecompositionofthegutbacterialcommunityoftheAmericanminkusinghigh-throughputsequencingtechnology. Methods FreshfecalsampleswerecollectedfromtenhealthyadultAmericanminkslivingincaptivity.DNAwasextractedusingtheQiagenStoolDNAextractionkitandtheV3-V4regionofthe16SrRNAgenewasamplifiedwithbarcodedprimers.ThelibrariesweresequencedontheIlluminaMiSeqplatform,generatingpaired-endreadsof300bp.ThequalityofthereadswasassessedusingFastQCandfilteredusingTrimmomatic.ThereadswerethenmergedusingFLASH,andchimericsequenceswereremovedusingUCHIME.Theremainingsequenceswereclusteredintooperationaltaxonomicunits(OTUs)usingtheUPARSEpipeline.TaxonomicclassificationwasperformedusingtheRDPBayesianclassifieragainsttheSILVAdatabase. Results Atotalof713,462high-qualitysequenceswereobtained,withanaverageof71,346sequencespersample.Therarefactioncurvesapproachedsaturation,indicatingthatthesequencingdepthwassufficienttocapturemostofthebacterialdiversityineachsample.Weidentifiedatotalof1,015OTUs,withanaverageof294OTUspersample.ThephylaFirmicutes,Bacteroidetes,andProteobacteriawerethemostabundant,accountingfor40.2%,30.8%,and19.6%ofthetotalreads,respectively.Atthegenuslevel,themostabundanttaxawereBacteroides(30.3%),Lactobacillus(16.8%),andRuminococcus(11.5%).Thealphadiversityindicesshowedthatthebacterialrichnessanddiversitywererelativelyhighintheminkgutmicrobiota,withanaverageShannonindexof4.3andanaverageChao1indexof436.ThebetadiversityanalysisbasedonBray-Curtisdissimilarityshowedthatthegutbacterialcommuniti
