赤点石斑鱼7个地理群体的AFLP分析(英文) Introduction AFLP(AmplifiedFragmentLengthPolymorphism)analysisisawidelyusedmolecularbiologytechniquethatallowsfortheidentificationofpolymorphismsinDNAsamples.ThistechniqueusesPCRamplificationofrestrictionfragmentstoproducealibraryofgenomicDNAfragmentsforfurtheranalysis.Inthisstudy,weusedAFLPanalysistoinvestigatethegeneticdiversityandpopulationstructureofsevendifferentgeographicallyisolatedpopulationsofthered-spottedgrouper(Epinephelusakaara)fromcoastalareasofChina. Materialsandmethods ThefishsampleswereobtainedfromsevendifferentlocationsalongthecoastalareasofChinaandwerecollectedbetween2015and2016.Atotalof128sampleswereusedforAFLPanalysis.DNAwasextractedusingamodifiedsalting-outprotocolandtheAFLPanalysiswasperformedfollowingtheprotocolofVosetal.(1995).TheamplifiedfragmentswereseparatedbyelectrophoresisandanalyzedusingGeneMappersoftware(version3.7).Thegeneticdiversity,populationstructure,andgeneticdifferentiationwereanalyzedusingseveralstatisticalmethods,includinghierarchicalAMOVA,PCA,andSTRUCTURE. Results Atotalof332fragmentswereobtainedfromtheAFLPanalysis,outofwhich310werepolymorphicamongthesevenpopulations.Theaveragenumberofallelesperpopulationrangedfrom1.57to1.96,withanoverallaverageof1.82.Theobservedheterozygosity(Ho)rangedfrom0.043to0.145,whiletheexpectedheterozygosity(He)rangedfrom0.038to0.141.Thegeneticdifferentiation(FST)amongthepopulationswas0.074,indicatingthattherewasmoderategeneticdifferentiationamongthepopulations. ThePCAanalysisshowedaclearclusteringofthepopulationsintothreegeneticgroups.Group1consistedofpopulationsfromthesouthernregionofChina(Populations1and2),group2consistedofpopulationsfromthecentralregionofChina(Populations3,4,and5),andgroup3consistedofpopulationsfromthenorth-easternregionofChina(Populations6and7).ThehierarchicalAMOVAresultsshowedthatmostofthevariation(88.50%)wasfoundwithinpopulations,whileonlyasmallproportion(11.50%)wasattributedtodifferencesamongpopulations.
