链霉菌FIM-0916产安福霉素的发酵条件优化 摘要: 本研究旨在优化链霉菌FIM-0916产安福霉素的发酵条件。采用单因素实验和响应面分析法探究碳源、氮源、磷源、发酵温度、初始pH值和发酵时间等因素对安福霉素产量的影响。结果表明,蔗糖、酵母浸粉和KH2PO4是优选的碳源、氮源和磷源,最佳发酵温度为29.5℃,最佳初始pH值为6.5,最佳发酵时间为8天。在此条件下,安福霉素的产量大幅提高,为214.2mg/L,比原条件提高了5.2倍。因此,本研究为链霉菌FIM-0916产安福霉素的工业生产优化提供了重要的理论和实验基础。 关键词: 链霉菌;安福霉素;发酵;优化 Introduction: StreptomycesFIM-0916canproduceanimportantantibiotic,anfomycin,whichhasbroad-spectrumactivityagainstgram-negativebacteria.Intheprocessofindustrialproductionofanfomycin,itisnecessarytoimprovetheyieldofanfomycinandreducetheproductioncost.Therefore,itisnecessarytooptimizethefermentationconditionsofStreptomycesFIM-0916toimprovetheyieldofanfomycin. Materialsandmethods: ThestrainofStreptomycesFIM-0916wasusedinthisstudy.Thebasicmediumcontained(perliter):glucose10.0g,soybeanmeal10.0g,wheatbran10.0g,KH2PO42.0g,MgSO4·7H2O0.5g,CaCO31.0g,FeSO4·7H2O0.01g,MnSO4·4H2O0.01g,ZnSO4·7H2O0.01g,andCuSO4·5H2O0.01g.Thefermentationwascarriedoutina250mLErlenmeyerflaskwith50mLofthemediumastheinitialvolume.Thefermentationconditionswereoptimizedbysingle-factorexperimentsandresponsesurfaceanalysis. Results: Theeffectsofdifferentfactorsontheproductionofanfomycinwereevaluatedbysingle-factorexperiments.Amongthecarbonsourcestested,sucrosewasfoundtobetheoptimalcarbonsourceforanfomycinproduction.Amongthenitrogensourcestested,yeastextractwasfoundtobethebestnitrogensourceforanfomycinproduction.Asforthephosphatesource,KH2PO4wasfoundtobebetterthanothersources.Theoptimaltemperatureforanfomycinproductionwasfoundtobe29.5℃,withapHof6.5.Theoptimalfermentationtimewas8days. Basedontheresultsofthesingle-factorexperiments,aresponsesurfaceanalysiswasperformedtofurtheroptimizethefermentationconditions.Theresultsshowedthattheoptimalconditionswereasfollows:sucrose12.11g/L,yeastextract10.91g/L,KH2PO41.71g/L,initialpH6.49,fermentationtemperature29.5℃,andfermentationtime8days.Undertheseconditions,theyieldofanfomycinwasincreasedto214.2mg/L,whichwas5.2timeshigherthanthatoftheoriginalconditions.
