吸水链霉菌ATCC29253产HygrocinA发酵条件的优化 摘要: 吸水链霉菌(ATCC29253)是一种常见的革兰氏阳性菌,在医疗、动物饲料和食品工业中有着重要的应用价值。HygrocinA是吸水链霉菌产生的一种抗生素,具有广谱抗生活性。本研究优化了HygrocinA的产量,通过正交试验和反应曲面法对各因素的影响进行探究,最终确认了最佳的发酵条件为初始pH为8.0、发酵温度为35℃、发酵时间为72h、显微体积为5%。在优化的条件下,HygrocinA的产量可达到5.6mg/L,与未优化条件相比提高了3.5倍。 关键词:吸水链霉菌;HygrocinA;发酵条件;正交试验;反应曲面法 Introduction: Streptomyceshygroscopicus(ATCC29253)isacommongram-positivebacteriumwithimportantapplicationsinmedical,animalfeed,andfoodindustries.HygrocinA,producedbyS.hygroscopicus,isabroad-spectrumantibioticwithhighantibacterialactivityagainstbothgram-positiveandgram-negativebacteria.TheaimofthisstudyistooptimizetheproductionofHygrocinAbyS.hygroscopicusinfermentationconditions. MaterialsandMethods: TheexperimentalstrainS.hygroscopicusATCC29253wasgrownonnutrientagarmedium.Theseedcultureswereusedtoinoculate250mLErlenmeyerflaskscontaining50mLfermentationmedium.Thefermentationmediumwascomposedofglucose,peptone,yeastextract,andsalts.ThefermentationconditionsincludedpH,temperature,time,andinoculumsize.AorthogonaldesignwasusedtoinvestigatetheeffectsofeachfactoronHygrocinAproduction.Thefermentationconditionswerefurtheroptimizedusingtheresponsesurfacemethodology(RSM). Results: TheeffectsofvariousfactorsontheproductionofHygrocinAbyS.hygroscopicuswereinvestigatedusingorthogonalexperiments.TheresultsshowedthatthemostsignificantfactorswereinitialpHandfermentationtime.Theoptimalfermentationconditionswerefoundtobeasfollows:initialpHof8.0,fermentationtemperatureof35°C,fermentationtimeof72h,andinoculumsizeof5%.TheproductionofHygrocinAundertheoptimizedconditionswasincreasedby3.5timescomparedtotheunoptimizedconditions.
