Introduction: Thebroadmoth,Helicoverpaarmigera,isamajoragriculturalpestthataffectsnumerouscrops,includingcotton,corn,soybeans,andvegetables.Todevelopeffectivepestmanagementstrategies,itisimportanttounderstandthemechanismsbehindtheinsect'sbehaviors,particularlyitsattractiontospecificodors.IthasbeenobservedthattheFas-Chaibuhuanproteinsareinvolvedinthematingbehaviorofthemalemoth,indicatingtheirpotentialastargetsforpestcontrol.Therefore,thisstudyfocusedoncloningandexpressingthegenesencodingtheseproteins. Methodology: 1.IdentificationofFas-Chaibuhuangenes:GenomicandtranscriptomicdataofH.armigerawereanalyzedtoidentifyputativeFas-Chaibuhuangenes.Multiplesequencealignmentandphylogeneticanalysiswereconductedtodeterminetheirevolutionaryrelationships. 2.Genecloning:Polymerasechainreaction(PCR)amplificationwasperformedusingspecificprimersdesignedbasedontheidentifiedFas-Chaibuhuangenes.Theamplifiedfragmentswerethenclonedintoasuitableexpressionvector. 3.Expressioninaheterologoussystem:TherecombinantvectorscontainingtheFas-ChaibuhuangenesweretransformedintoEscherichiacoliorothersuitablehostcells.Proteinexpressionwasinduced,andtherecombinantproteinswerepurifiedusingaffinitychromatography.
