MaterialsandMethods Reagents TheFFXTCwasobtainedfromaChinesemedicinepharmacyandwasdissolvedindistilledwaterforinvitroexperiments.Collagen,arachidonicacid(AA),thrombin,andADPwereusedtostimulateplateletaggregation.Thethrombingenerationassaykitwasusedtodeterminethrombingenerationinplasma.Theactivatedpartialthromboplastintime(APTT)andprothrombintime(PT)wereperformedusingcommercialkits. Plateletaggregationassay Platelet-richplasma(PRP)waspreparedfromhumanbloodanddifferentconcentrationsofFFXTCwereaddedtoPRP.Plateletaggregationinducedbydifferentagonistswasmeasuredusingaturbidimetricmethod. Thrombingenerationassay ThrombingenerationwasmeasuredinhumanplasmatreatedwithdifferentconcentrationsofFFXTCusingthethrombingenerationassaykitaccordingtothemanufacturer’sinstructions. PTandAPTTassay PTandAPTTweredeterminedinhumanplasmatreatedwithdifferentconcentrationsofFFXTC. Results FFXTCinhibitedplateletaggregationinducedbycollagen,AA,thrombin,andADPinaconcentration-dependentmanner.Thiseffectwasmorepotentoncollagen-inducedplateletaggregationthanonotherstimuli.FFXTCdose-dependentlyprolongedAPTTandPT,indicatingitseffectontheintrinsicandextrinsiccoagulationpathways.FFXTCalsoinhibitedthrombingenerationinplasma.
