TranscriptionalregulationofAdiposegene TheAdiposegeneisexpressedinavarietyoftissues,includingadiposetissue,liver,muscle,andbrain.TheexpressionofAdiposeisregulatedbyseveraltranscriptionfactors,suchasperoxisomeproliferator-activatedreceptorgamma(PPARγ),CCAAT/enhancer-bindingproteinalpha(C/EBPα),andsterolregulatoryelement-bindingprotein1c(SREBP-1c).Amongthesefactors,PPARγisthemostimportantregulatorofadipogenesisandisessentialforthedifferentiationofpreadipocytesintomatureadipocytes.PPARγbindstotheAdiposepromoterandactivatesitstranscription,therebypromotingadipocytedifferentiationandlipidaccumulation. InadditiontoPPARγ,C/EBPαalsoplaysacriticalroleinadipogenesisbyactivatingtheexpressionofPPARγandotheradipogenicgenes.C/EBPαbindstotheAdiposepromoterandenhancesitsactivity,therebycontributingtotheregulationoflipidmetabolismandinsulinsensitivity.SREBP-1cisanotherregulatorofAdiposegeneexpression,whichisinvolvedintheregulationofthebiosynthesisoffattyacidsandtriglycerides.SREBP-1cbindstotheAdiposepromoterandactivatesitstranscription,therebypromotinglipidsynthesisandstorage.
