Adipose基因转录调控及功能研究的中期报告 【Abstract】 Adiposetissueisanimportantorganinvolvedinenergymetabolism,endocrineregulation,andimmunefunction.Dysregulationofadiposetissuedevelopmentandfunctioncanleadtoobesityandrelatedmetabolicdisorders.Adiposetissuedevelopmentandfunctionareregulatedbycomplexgeneexpressionnetworks,includingtranscriptionfactors,non-codingRNAs,andepigeneticregulators.Inthisstudy,weaimtoinvestigatethetranscriptionalregulationandfunctionalrolesoftheAdipose(Adip)gene,whichencodesatranscriptionfactorthatwaspreviouslyidentifiedasaregulatorofadipogenesisandlipidmetabolism. 【Introduction】 Adiposetissueisacomplexanddynamicorganthatplaysacriticalroleinenergyhomeostasis,endocrinesignaling,andimmunefunction.Dysregulationofadiposetissuedevelopmentandfunctioncanleadtoobesity,insulinresistance,type2diabetes,andothermetabolicdisorders.Adiposetissuedevelopmentandfunctionareregulatedbyacomplexinterplayofgeneticandenvironmentalfactors,includingtranscriptionfactors,non-codingRNAs,andepigeneticregulators.Adipose-specifictranscriptionfactorshavebeenidentifiedaskeyregulatorsofadiposetissuedevelopment,differentiation,andfunction.Amongthese,theAdiposegene(Adip)hasbeenshowntoplayacriticalroleinadipocytedifferentiationandlipidmetabolism. 【Methods】 ToinvestigatethetranscriptionalregulationandfunctionalrolesoftheAdipgene,weusedacombinationofmoleculargenetics,bioinformatics,andfunctionalassays.WegeneratedstablecelllinesoverexpressingAdipandperformedRNA-SeqandChIP-SeqanalysestoidentifythedownstreamtargetsofAdipandthegenomicregionsboundbyAdip.Wealsoperformedfunctionalassays,includingcellproliferation,adipocytedifferentiation,andlipidmetabolismassays,toinvestigatetheroleofAdipinadiposetissuedevelopmentandfunction. 【Results】 OurRNA-SeqanalysisidentifiedasetofadipogenicandlipogenicgenesthataredifferentiallyregulatedbyAdip.ChIP-SeqanalysisrevealedthatAdipbindstothepromoterregionsofthesetargetgenes,suggestingthatAdipdirectlyregulatestheirexpression.FunctionalassaysshowedthatAdipoverexpressionpromo
