ddPCR检测乳腺癌新辅助化疗前后HER2变化的临床意义 Title:ClinicalSignificanceofddPCRinDetectingHER2ChangesbeforeandafterNeoadjuvantChemotherapyforBreastCancer Abstract: Breastcanceristhemostcommoncanceramongwomenworldwide,andHER2(humanepidermalgrowthfactorreceptor2)isanimportantbiomarkerassociatedwithtumoraggressivenessandresponsetotargetedtherapies.Neoadjuvantchemotherapy,administeredpriortosurgery,hasbecomethestandardtreatmentforlocallyadvancedbreastcancer.ThispaperaimstoexploretheclinicalsignificanceofdropletdigitalPCR(ddPCR)indetectingHER2changesbeforeandafterneoadjuvantchemotherapyforbreastcancer,includingitsroleinpredictingtreatmentresponseandguidingtherapeuticdecision-making. Introduction: Breastcancerisacomplexdiseasewithhighheterogeneity.AccurateassessmentofHER2statusiscrucialfordeterminingappropriatetargetedtherapy.Traditionalmethods,suchasimmunohistochemistry(IHC)andfluorescenceinsituhybridization(FISH),havelimitationsintermsofsubjectivity,inter-laboratoryvariability,andinsufficientsensitivity.DropletdigitalPCR(ddPCR)isanovelmoleculartechniquethatallowsabsolutequantificationofDNAtargetswithhighprecisionandsensitivity.ThispaperaimstohighlighttheclinicalsignificanceofddPCRindetectingchangesinHER2statusbeforeandafterneoadjuvantchemotherapyforbreastcancer. Methods: AsystematicreviewoftheliteraturewasconductedtoidentifyrelevantstudiesfocusingonHER2assessmentusingddPCRinbreastcancerpatientsbeforeandafterneoadjuvantchemotherapy.ThesearchincludedstudiespublishedinelectronicdatabasessuchasPubMed,Embase,andCochraneLibrary.StudiesreportingontheclinicalsignificanceofHER2changesdetectedbyddPCRwereincludedforanalysis. Results: SeveralstudieshaveinvestigatedtheuseofddPCRindetectingHER2changesduringneoadjuvantchemotherapyforbreastcancer.TheresultssuggestthatHER2geneamplificationmayvaryaftertreatment,leadingtochangesinHER2status.ThisdynamicchangeinHER2statusmayhaveimportantclinicalimplications,asitcanimpacttreatmentresponseandsubsequenttherapeuticdecisions.ddPCRhasdemonstratedsuperiorityindete
