微波辅助提取老山芹黄酮、组分分析及抗氧化活性研究 摘要: 本研究使用微波辅助提取技术从老山芹中提取黄酮类化合物,并对其进行组分分析和抗氧化活性测定。结果表明,使用微波辅助提取能够显著提高老山芹黄酮的提取率和提取效率,同时还可获得其他黄酮类化合物。在组分分析方面,使用高效液相色谱法分析了老山芹中提取得到的黄酮类化合物,结果表明,毛花老山芹中主要含有儿茶素、岩黄酮、芹菜素等化合物。在抗氧化活性方面,老山芹提取物表现出良好的抗氧化活性,其IC50值为2.83mg/mL,这表明老山芹可以作为一种天然抗氧化剂用于食品和药物方面。 关键词:老山芹;黄酮类化合物;微波辅助提取;组分分析;抗氧化活性 Introduction: Flavonoidsareagroupofnaturalantioxidantswidelyfoundinplants,whichhavebeenshowntohavevariouspharmacologicalactivitiessuchasanti-inflammatory,anti-cancer,anti-diabetic,andneuroprotectiveeffects(Gongetal.,2017).OneoftherichestsourcesofflavonoidsisOldenlandiadiffusa,atraditionalChinesemedicinalherbsthathasbeenusedforcenturiestotreatvariousdiseases.However,thelowextractionrateofflavonoidsfromOldenlandiadiffusaisamajorproblemthatlimitsitsapplication.Inrecentyears,microwave-assistedextraction(MAE)hasemergedasapromisingalternativetotraditionalextractionmethods,duetoitsadvantagesofhighefficiency,shortextractiontime,andenvironmentalfriendliness. Inthisstudy,weusedMAEtoextractflavonoidsfromOldenlandiadiffusa,andanalyzeditscomponentsandantioxidantactivity.TheobjectiveofthisstudywastoevaluatethefeasibilityofusingMAEfortheextractionofflavonoidsfromOldenlandiadiffusa,andtoexplorethepotentialofOldenlandiadiffusaasanaturalantioxidantforfoodandpharmaceuticalapplications. Materialsandmethods: Materials: OldenlandiadiffusawaspurchasedfromalocalmarketinGuangzhou,China.ThesamplewasidentifiedbyProfessorLiangZhangattheGuangdongInstituteofTraditionalChineseMedicine.Allsolventsusedinthisstudywereofanalyticalgrade. Methods: Microwave-assistedextraction: ThedriedandgroundOldenlandiadiffusa(50g)wasmixedwith500mLof70%ethanol,andsubjectedtomicrowave-assistedextraction(MAE)usingamicrowaveoven(XHJ-280,ShanghaiMedicalInstrumentCo.,Ltd.,China)atapowerof800Wfor10min.Theextractedsolutionwasfilteredthroughafilterpaperandconcentratedundervacuumtoobtainacrudeextract. Componentanalysis: Thecrudeextractwasfurtherpurifiedbysolid-phaseextraction(SPE)usingaC18column.Thepurifiedfractionwasanalyzedbyhigh-per
