Keywords:Perillaseedoil,responsesurfacemethodology,anti-inflammatoryactivity,bioactivecompounds,optimization. 1.Introduction Perillafrutescens,awidelygrownherbinAsia,isknownforitsnumerousmedicinalproperties.Theoilextractedfromitsseeds,commonlyknownasPerillaseedoil(PSO),isrichinpolyunsaturatedfattyacids(PUFAs),suchasalpha-linolenicacid(ALA).Theanti-inflammatoryactivityofPSOhasattractedconsiderableattention,makingitapotentialtherapeuticagentforinflammatorydiseases.However,thequalityandcompositionofPSOcanvarydependingonvariousfactors,includingcultivationconditionsandextractionmethods.Therefore,itisessentialtooptimizethepreparationprocesstoenhanceitsbioactiveproperties. 2.MaterialsandMethods 2.1ExperimentalDesign Responsesurfacemethodology(RSM)wasemployedtooptimizethepreparationprocessofPSO.Factorssuchasextractiontemperature,extractiontime,andsolvent-to-seedratiowereselectedasindependentvariables.Thedependentvariablesincludedoilyield,ALAcontent,andtotalphenoliccontent.ABox-Behnkendesign(BBD)wasusedfortheexperimentaldesign,andtheresponsedatawerefittedtosecond-orderpolynomialequations. 2.2OilExtractionandAnalysis PerillafrutescensseedswerecollectedfromChangbaiMountainanddried.PSOwasobtainedbysolventextractionusingaSoxhletapparatus.Theoilyieldwasdeterminedgravimetrically.ThecontentofALAwasdeterminedusinggaschromatography,andthetotalphenoliccontentwasmeasuredusingtheFolin-Ciocalteumethod. 2.3Anti-inflammatoryActivityEvaluation Theanti-inflammatoryactivityofPSOwasevaluatedusinginvitroandinvivomodels.Theinvitroassaysincludedtheinhibitionof
