芹菜素对非洲爪蟾卵母细胞体外成熟的影响及其作用机制研究 Abstract: Apigeninisaflavonoidcompoundfoundinceleryandmanyotherplants,havingdiversebiologicalactivities.Inthisstudy,weinvestigatedtheeffectofapigeninoninvitromaturationofAfricanclawedfrog(Xenopuslaevis)oocytesandexploredtheunderlyingmechanisms.Ourresultsshowedthatapigeninatconcentrationsof10-50μMsignificantlyinhibitedthegerminalvesicle(GV)breakdown,firstpolarbodyextrusion,andmaturationofoocytesinadose-dependentmanner.Moreover,apigenintreatmentresultedinadecreaseintheexpressionofmaturation-promotingfactor(MPF)subunitCyclinB1andCdk1inoocytes.Wealsofoundthatapigenininhibitedoocytematurationbyaffectingtheactivityofthemitogen-activatedproteinkinase(MAPK)pathway,asevidencedbydecreasedphosphorylationlevelsofMAPKkinases(MEK)andextracellularsignal-regulatedkinase(ERK).Furthermore,apigenintreatmentresultedinDNAdamageandtheactivationofp53signalingpathway,whichmayalsoplayaroleinapigenin-inducedinhibitionofoocytematuration.Inconclusion,ourfindingssuggestthatapigeninimpairsoocytematurationthroughinhibitionoftheMPFcomplexandtheMAPKsignalingpathway,aswellasinducingDNAdamageandactivatingp53signalingpathway. Introduction: Apigenin(4',5,7-trihydroxyflavone)isaflavonoidcompoundthatbelongstotheflavonesubclassofflavonoids,whicharefoundinavarietyoffruits,vegetables,andherbs,includingcelery,parsley,chamomile,andthyme.Apigeninhasbeenshowntopossessawiderangeofbiologicalactivities,includingantioxidant,anti-inflammatory,anti-tumor,andanti-proliferativeeffects.Previousstudieshavedemonstratedthatapigeninisinvolvedinmanyimportantcellularprocesses,suchascellcycle,apoptosis,andDNArepair,throughregulatingvarioussignalingpathways.
