树鼩组织的全长转录组与组织表达谱分析 Title:TranscriptomeandTissueExpressionProfilingofTarsiers:AComprehensiveStudyontheTreeShrewOrganization Abstract: Understandingthetranscriptomeandtissueexpressionprofilesofanorganismiskeytounravelingitsfunctionalgenomics.Inthisstudy,weinvestigatethefull-lengthtranscriptomeandtissueexpressionpatternsoftreeshrews(Tupaiabelangeri),asmallarborealmammalbelongingtotheScandentiaorder.Throughthiscomprehensiveanalysis,weaimtogaininsightsintothegeneticandmolecularbasisofthetreeshreworganization. Introduction: Thetreeshrewisanemergingmodelorganismduetoitsclosephylogeneticrelationshipwithprimatesandasharedevolutionaryhistory.Recentadvancesinhigh-throughputsequencingtechnologieshaveprovidedanopportunitytoexplorethetranscriptomeandtissue-specificexpressionpatternsindetail.Thisstudyaimstounravelthegeneticandmolecularmechanismsgoverningthetreeshreworganizationbyanalyzingthefull-lengthtranscriptomeandtissueexpressionprofiles. Methods: RNAsampleswereextractedfromvarioustissuesofadulttreeshrews,includingthebrain,liver,heart,kidney,andspleen.ThetotalRNAwasenrichedformRNA,andfull-lengthcDNAlibrarieswereconstructedusingamodifiedprotocol.Thelibrariesweresequencedusinganext-generationsequencingplatformtoobtainhigh-qualityreads.Bioinformaticsanalysis,includingtranscriptomeassembly,annotation,anddifferentialexpressionanalysis,wasperformedusingstate-of-the-arttoolsanddatabases. Results: Thetranscriptomeassemblyyieldedacomprehensivecatalogoftranscripts,includingbothknownandnovelcodingandnon-codingtranscripts.Theannotationofthetranscriptomerevealedthefunctionalannotationandclassificationofprotein-codinggenesandnon-codingRNAs.Thedifferentialexpressionanalysisprovidedinsightsintotissue-specificgeneexpressionpatterns,revealinggenesthatarehighlyexpressedandgenesthatarespecifictocertaintissues.Thetissueexpressionprofilingunveiledtheuniqueexpressionsignaturesofvarioustissues,highlightingtheirfunctionalspecializationandpotentialroleinphysiologicalprocesses.
