基于形态学标记和SSR标记的荷花品种遗传多样性分析 Title:GeneticDiversityAnalysisofLotusVarietiesBasedonMorphologicalandSSRMarkers Abstract: Lotus(Nelumbogenus)isanimportantaquaticplantwithhighculturalandeconomicvalue.Thegeneticdiversityanalysisoflotusvarietiesplaysacrucialroleintheconservationandutilizationofthisspecies.Inthisstudy,weaimedtoinvestigatethegeneticdiversityoflotusvarietiesusingmorphologicalandSSRmarkers.AtotalofXXlotusvarietieswerecollectedandcharacterizedbasedonelevenmorphologicaltraits.Additionally,SSRmarkerswereemployedtoassessthemoleculargeneticdiversity. Introduction: Lotus,belongingtotheNelumbogenus,iswidelydistributedinvariousregionsofAsia,Africa,andNorthAmerica.Itisnotonlyabeautifulaquaticflowerbutalsoanimportantcropandmedicinalplant.Traditionalmethodsforstudyingthegeneticdiversityoflotusvarietiesmainlyrelyonmorphologicalobservationandevaluation.However,thesemethodshavelimitationsinaccuracyandobjectivity.Withthedevelopmentofmolecularbiology,SSRmarkershavebeenwidelyusedforgeneticdiversityanalysisduetotheirco-dominantinheritance,abundance,andhighpolymorphism. Methods: 1.Collectionandcharacterizationoflotusvarieties: -AtotalofXXlotusvarietieswerecollectedfromdifferentregions. -Elevenmorphologicaltraits,includingpetalcolor,leafshape,andcarpelnumber,wererecordedandanalyzed. 2.ExtractionofgenomicDNAandSSRmarkeranalysis: -GenomicDNAwasextractedfromyoungleavesofeachlotusvariety. -AsetofpreviouslydevelopedSSRmarkersspecifictolotuswereselected. -PCRamplification,gelelectrophoresis,andvisualizationofSSRalleleswereperformed. -Thenumberofalleles,allelefrequency,polymorphisminformationcontent(PIC),andgeneticidentitywerecalculatedbasedontheSSRdata. Results: 1.Morphologicalcharacterization: -TheanalyzedmorphologicaltraitsshowedsignificantvariationamongtheXXlotusvarieties. -Petalcolorexhibitedthehighestdiversity,followedbyleafshapeandcarpelnumber. 2.SSRmarkeranalysis: -XXSSRmarkerswereusedforgeneticdiversityassessment. -AtotalofXXallelesweredetected,withanaverageofXXallelespermarker.
