Introduction: Goat'smilkisavaluablesourceofnutrients,butitcontainsallergenicproteinssuchasβ-lactoglobulin(BLG),whichcaninduceallergicreactionsinsusceptibleindividuals.KnockingouttheBLGgeneingoatswouldeliminateorreducetheproductionofBLG,resultinginhypoallergenicmilk.Ontheotherhand,humanlactoferrin(hLF)isamultifunctionalproteinwithvarioushealthbenefits.IntroducingthehLFgeneintothegoatgenomewouldenhancethenutritionalvalueofgoat'smilk.TALENsarecustomizablenucleasesthathavebeenwidelyusedforprecisegenomeeditinginvariousorganisms,includinglivestockspecies.Inthisstudy,weaimedtoutilizeTALENstoknockouttheBLGgeneandknockinthehLFgeneingoats. Methods: DesignandconstructionofTALENvectors:TALENsweredesignedtotargetspecificsiteswithintheBLGgeneusingamodularassemblyapproach.TheTALENpairswereclonedintoexpressionvectorsandthenvalidatedfortheiractivityinculturedgoatsomaticcells. Embryomicroinjection:FertilizedgoatembryoswerecollectedandsubjectedtomicroinjectionwithTALENmRNA,followedbyinvitroculturetoallowfordevelopmenttotheblastocyststage.GenomicDNAwasisolatedfromtheresultingblastocystsforgenotypinganalysis.
