ARK5在肝细胞肝癌对多柔比星耐药性中的作用机制的研究 Title:MechanismsofARK5inMultidrugResistanceinHepatocellularCarcinoma Abstract: Hepatocellularcarcinoma(HCC)isoneofthemostprevalentandaggressiveformsoflivercancer,characterizedbyhighratesofchemoresistanceandpoorprognosis.Thedevelopmentofmultidrugresistance(MDR)posesasignificantchallengetotheeffectivetreatmentofHCC.ThispaperaimstoinvestigatetheroleofARK5inMDRinHCCandexploreitsunderlyingmechanisms.UnderstandingthemechanismsbywhichARK5contributestoMDRinHCCisessentialforthedevelopmentoftargetedtherapiesandimprovedpatientoutcomes. 1.Introduction: Hepatocellularcarcinoma(HCC)isthemostcommontypeofprimarylivercancerandisresponsibleforasignificantnumberofcancer-relateddeathsworldwide.Despiteadvancesindiagnosisandtreatment,HCCremainshighlyresistanttoconventionalchemotherapyandtargetedtherapy,leadingtopoorpatientsurvivalrates.Multidrugresistance(MDR)isamajorobstacleintheeffectivetreatmentofHCC,necessitatingextensiveresearchtoidentifynoveltherapeutictargetsandelucidatetheunderlyingmechanisms. 2.ARK5anditsroleinHCC: ARK5,alsoknownasAMPK-relatedkinase5,isaproteinkinaseinvolvedinvariouscellularprocesses,includingcellproliferation,apoptosis,andenergymetabolism.EmergingevidencesuggeststhatARK5playsacrucialroleincancerprogression,includingHCC.StudieshaveshownthatARK5expressioniselevatedinHCCtissuescomparedtonormallivertissues,anditsoverexpressionisassociatedwithapoorprognosis.Moreover,increasedARK5expressionisfrequentlyobservedinHCCpatientswhodevelopchemoresistance.ThesefindingssuggestthatARK5maycontributetothedevelopmentofMDRinHCC. 3.SignalingpathwaysregulatedbyARK5inMDR: SeveralsignalingpathwayshavebeenidentifiedasdownstreamtargetsofARK5thatmaymediateitsroleinMDRinHCC.OnesuchpathwayisthePI3K/Akt/mTORpathway,whichisknowntoregulatecellsurvival,growth,anddrugresistance.ARK5hasbeenshowntoactivatethePI3K/Akt/mTORpathway,leadingtoincreasedproliferationandsurvivalofcancercells.Additionally,ARK5hasbeenfoundtoactivatetheWnt/β-cateninsignalingpathway,whichisinvolve
