利用代谢组学方法筛选肝外胆管癌血浆诊断标志物的初步研究的中期报告 摘要: 肝外胆管癌是一种恶性肿瘤,其早期诊断能够显著提高患者的生存率。本研究旨在利用代谢组学方法筛选肝外胆管癌血浆诊断标志物,以期为肝外胆管癌的早期诊断提供新的思路和方法。我们采集了10例肝外胆管癌患者和10例健康志愿者的血浆样本,利用超高效液相色谱-高分辨质谱(UPLC-HRMS)技术进行代谢组学分析,并采用多变量统计方法对数据进行分析。结果显示,肝外胆管癌患者的血浆代谢谱与健康志愿者明显不同,共筛选出了26个代谢物可能与肝外胆管癌相关,其中10个代谢物具有较高的区分度。这些代谢物主要涉及葡萄糖代谢、脂质代谢、氨基酸代谢等方面,这些代谢变化可能与肝外胆管癌的病理生理过程密切相关。本研究为肝外胆管癌早期诊断提供了一些新的诊断标志物,但还需要进一步研究和验证。 关键词:代谢组学;肝外胆管癌;血浆;诊断标志物;多变量分析 Abstract: Extrahepaticcholangiocarcinomaisamalignanttumor,andearlydiagnosiscansignificantlyimprovethesurvivalrateofpatients.Theaimofthisstudyistoscreenfordiagnosticbiomarkersofextrahepaticcholangiocarcinomainplasmausingmetabolomicsmethodstoprovidenewideasandmethodsfortheearlydiagnosisofthisdisease.Wecollectedplasmasamplesfrom10extrahepaticcholangiocarcinomapatientsand10healthyvolunteers,performedmetabolomicsanalysisusingultra-highperformanceliquidchromatography-highresolutionmassspectrometry(UPLC-HRMS)technology,andanalyzedthedatausingmultivariatestatistics.Theresultsshowedthattheplasmametabolicprofileofpatientswithextrahepaticcholangiocarcinomawassignificantlydifferentfromthatofhealthyvolunteers,andatotalof26metabolitesthatmayberelatedtoextrahepaticcholangiocarcinomawerescreenedout,including10metaboliteswithhighdiscrimination.Thesemetabolitesmainlyinvolveglucosemetabolism,lipidmetabolism,aminoacidmetabolism,andotheraspects,andthesemetabolicchangesmaybecloselyrelatedtothepathologicalandphysiologicalprocessesofextrahepaticcholangiocarcinoma.Thisstudyprovidessomenewdiagnosticbiomarkersfortheearlydiagnosisofextrahepaticcholangiocarcinoma,butfurtherresearchandvalidationareneeded.
