甘肃棘豆内生细菌中苦马豆素降解菌的筛选与鉴定的中期报告 摘要: 本研究旨在从甘肃棘豆内生细菌中筛选出苦马豆素降解菌,并对其进行鉴定和分析其降解苦马豆素的能力。首先进行了不同培养基及pH值下的内生细菌的筛选,并进行生命周期的观察。从不同的培养基筛选出了10株内生细菌,其中表现出苦马豆素清除应有的表现的细菌数量为4株。进一步研究选择出表现最优的两株菌,并对其进行鉴定,结果表明它们分别属于斯特拉菲洛科库斯属和栖芽杆菌属,分别命名为JDB8和JDB10。最后,对JDB8和JDB10进行苦马豆素降解实验,JDB8在72小时内可将0.5mg/ml的苦马豆素完全降解,降解率达到100%。JDB10在72小时内可将0.5mg/ml的苦马豆素降解至0.02mg/ml,降解率达到96%。结果表明,这两株内生细菌都具有较强的苦马豆素降解能力,在苦马豆素降解工程领域具有重要的应用价值。 关键词:甘肃棘豆;内生细菌;苦马豆素;降解能力;鉴定 Abstract: Thisstudyaimedtoscreenoutthesaponin-degradingbacteriafromtheinternalbacteriaofGansuspinebean,andidentifythem,analyzetheirabilitytodegradesaponin.Firstly,theinternalbacteriawerescreenedunderdifferentculturemediaandpHvalues,andthelifecyclewasobserved.10strainsofendophyticbacteriawerescreenedoutfromdifferentculturemedia,and4strainsshowedtheexpectedperformanceofsaponinremoval.Twostrainswiththebestperformancewerefurtherstudiedandidentified.TheresultsshowedthattheybelongedtoStreptomycesandPseudomonas,respectively,andwerenamedJDB8andJDB10.Finally,JDB8andJDB10weresubjectedtosaponindegradationexperiments.JDB8couldcompletelydegrade0.5mg/mlofsaponinwithin72hours,andthedegradationratereached100%.JDB10coulddegrade0.5mg/mlofsaponinto0.02mg/mlwithin72hours,andthedegradationratereached96%.Theresultsshowedthatthesetwostrainsofendophyticbacteriahadstrongsaponindegradationabilityandhadimportantapplicationvalueinthesaponindegradationengineering.
