Interrelationshipamongthepolymorphismof(AC)ninthe i;,-endoftheARgene,theerythrocytealdosereductase activityandsusceptibilityofmicroangiopathyinthepatients withtype2diabetesmellitus Abstract ObjectiveTostudytheinterrelationshipamongthepolymorphismof(AC)ninthe 5'-endoftheARgene,theerythrocytealdosereductaseactivityandsi!sceptibility ofmicroangiopathyinthepatientswithtype2diabetesmellitus(DM).Methods Onehundredandfortycasesoftype2DMandforty-ninecontrolswereincluded inthisstudy.AllthesecasesweretheHannationalityofGuangxizhuang autonomousregion.ThesubjectsweredevidedintoDMwithoutdiabetic microangiopathy(NDC)(56cases),DMAPgrouy(84cases),andnormal controls(CON)(44cases),furthermore,accordingtothetypeofallele,thesubjects weredevidedintoZ+2,Z-2,bothZ+2andZ-2,neitherZ+2norZ-2(X/X).PCR wasperformedandPCRproductswereelectrophoresedwith8%formamid/urea gel.ARactivity(ARA)wasmeasuredbymodifiedSriratavamethod.The differencesofARAwerecomparedamongthesegroups.ResultsItshowed7types ofalleles(Z-6-Z+6).ThefrequencyofalleleZ+2wasmuchlowerinDMgroup thaninNDCgroupandCONgroup.ARAinDMAPgroupwassignificantlyhigher thanthatinNDCgroup,ARAinNDCgroupwassignificantlyhigherthanthatin CONgroup.AmongZ-2,Z+2,Z+2/Z-2andX/Xgroups.ARAwasthehighestin DMZ-2/Z-2groupandthelowestinDMZ十21Z+2group.ConclusionZ-2 .1。 allelemightbethesusceptiblegeneandZ+2allelemightbetheprotectedgene.
