浅谈动态观察柴胡皂苷a对实验性癫痫小鼠痫性发作的影响浅谈动态观察柴胡皂苷a对实验性癫痫小鼠痫性发作的影响【关键词】柴胡皂苷摘要:目的研究柴胡皂苷a(saikosaponina,SSa)对戊四氮(PTZ)诱导的小鼠痫性发作的影响,了解其药效特点。方法采用PTZ诱导小鼠急性痫性发作来制作癫痫模型,通过记录不同时间点小鼠痫性发作潜伏期和强直性惊厥率的变化,研究SSa抗小鼠PTZ致痫作用的动态特征。结果SSa腹腔注射30、60、90min组能显着延长阵发性痉挛潜伏期(P0.05),其中SSa90min组作用最为显着(P001)。在60、90、120min组中,SSa腹腔注射也能明显延长强直性惊厥潜伏期(P0.05),其中尤以90min组作用显着(P001);SSa90min组对过渡时间延长作用显着(P0.05);SSa30min、60min组能减少PTZ致痫小鼠强直性惊厥发作例数(P005),且以SSa60min组作用最为显着。结论SSa具有对抗小鼠PTZ致痫作用;腹腔注射SSa对PTZ诱导小鼠的痫性发作在120min内以60min、90min药效作用较强。关键词:实验性癫痫;柴胡皂苷a;PTZ;小鼠Abstract:ObjectiveTostudythedynamicvariationsoftheeffectofsaikosaponina(SSa)onepilepticmiceinducedbypentylenetetrazol(PTZ),andtounderstandtheeffectcharacteristicsofSSa.MethodWiththeepilepticmiceinducedbyPTZasanimalmodel.Inthedifferenttimepoint,thetentativeseizureincubationperiodandthecaseoftetanicconvulsionwererecordedtostudythecharacteristicsoftheantiepilepticeffectofSSaonmouseinducedbyPTZ.ResultsInthegroupof30min,60minand90min,theincubationperiodofcloniccrampwassignificantlyprolongedbySSa(P0.05),andinthegroupof90min,theeffectwasremarkable(P0.01).Also,intraperitonealinjectionofSSacouldprolongtheincubationperiodoftetanicconvulsioninthegroupof60min,90minand120min(P0.05).Theeffectwasespeciallyevidentinthegroupof90min(P0.01).Inthegroupof90min,thetransitiontimewasremarkablyprolongedbySSa(P0.05).InthegroupofSSa30minandSSa60min,SSacouldreducethetetanicconvulsionofepilepticmiceintroducedbyPTZ(P0.05),andtheeffectismoresignificantinthegroupof60minthanthatinthegroupof30min.Inthegroupof30minand60min,SSacouldprotecttheepilepticmiceintroducedbyPTZfromtetanicconvulsionmoresignificantlythanthoseinthegroupof90minand120min.ConclusionsSSacaninhibittheepilepticmiceinducedbyPTZ,SSacanprotectthemousefromtetanicconvulsion,andin2hours,theeffectwasmoreremarkableat60thmin,90thminthanothers.Keywords:epilepsy;Saikosaponina;PTZ;mice柴胡皂苷(saikosaponin)为中药柴胡的主要成分之一,其单体成分之一的柴胡皂苷a(SSa)有降血脂、抗渗出、抗肉芽肿和保护肝细胞等作用[1],对于慢性肾病的治疗也有较好疗效[2,3],但目前尚没有SSa抗癫痫作用的报道。不过已有不少证据表明柴胡及柴胡总皂苷具有镇静和抗实验性癫痫的作用[4,5]。在柴胡皂苷单体各成分中,为明确何种皂
