Aug.2009Volume6No.8(SerialNo.57)美中医学JournalofUS-ChinaMedicalScienceISSN1548-6648USA同种异体移植的骨髓间充质干细胞在椎间盘环境中存活及增殖能力的研究李华壮1方军1周跃2王卫东2(1.潍坊医学院第三附属医院山东青州262500;2.第三军医大学新桥医院重庆400038)摘要:目的观察同种异体移植的骨髓间充质干细胞(MSCs)在椎间盘环境中的存活及增殖能力。方法从30只日本大白兔中分离培养骨髓MSCsAd-EGFP标记后同种异体移植到椎间盘环境中术后1周、2周、4周、8周、12周取材Brdu处理后做石蜡切片荧光显微镜观察细胞存活并行Brdu免疫组化染色检测细胞增殖。结果术后1周、2周、4周、8周、12周标本内可观察到绿色荧光术后1周、2周、4周、8周、12周标本内均有Brdu免疫组化染色阳性细胞。结论自体移植的MSCs在椎间盘环境中能够存活并且增殖为MSCs作为椎间盘组织工程的种子细胞提供了实验依据。关键词:椎间盘;骨髓间充质干细胞;同种异体移植;存活;增殖TheviabilityandproliferationofallograftedmesenchymalstemcellsintheintervertebraldiscLIHua-zhuangFANGJunZHOUYueWANGWei-dongAbstract:ObjectiveToobservetheviabilityandproliferationofallograftedmessenchymalstemcellsintheintervertebraldisc.MethodsBonemesenchymalstemcellsisolatedfrom12JapanesewhiterabbitswerelabeledwithAd-EGFPandwereallograftedintolumbarintervertebraldiscofwhichthenucleuspulposushadbeenaspirated.Thelumbarspinewereextracted124812weeksaftertheoperationdealedwithBrdutheviabilityandproliferationofthecellswereassessedwithconsecutivesectionsusingfluorescentmicroscopeandimmunohistochcmistryofBrdu.ResultsFluorescencewasobservedinthespecimensfor124812weeksaftertheoperationandpositiveimmunohistochemicalstainingforBrduwasobservedinthespecimensfor124812weeksaftertheoperation.ConclusionAllograftedmesenchymalstemcellscansurviveandproliferateintheintervertebraldisc.ResultsfromourstudyprovideexperimentalevidenceforMSCsasseedingcellsofintervertebraldisctissueengi
