Introduction: Soybean(Glycinemax)isoneofthemostimportantcropsforbothfoodandoilproduction.However,theoccurrenceofrootrotdiseases,causedbyvariouspathogens,severelyimpactssoybeanproduction.Soil-bornepathogenssuchasFusariumspp.,Phytophthorasojae,Pythiumspp.,andRhizoctoniaspp.areknowntocausesoybeanrootrot.Conventionaldiagnosticmethods,includingmorphologicalidentificationandpathogenicitytests,havelimitationsintermsofaccuracyandspeed.Therefore,thedevelopmentofmoleculardetectiontechniquesholdsgreatpromiseinidentifyingthesepathogensandenhancingdiseasemanagementstrategies. MethodsandResults: 1.PathogenIsolationandMorphologicalIdentification: Infectedsoybeanrootswerecollectedfromdifferentregionsandplatedonselectivemediatoisolatetherootrotpathogens.Morphologicalcharacteristics,suchasmycelialgrowth,colonycolor,andsporemorphology,wereobservedtoidentifythepathogenstothegenusorspecieslevel.CompatibilitytestsandKoch'spostulateswereperformedtoconfirmpathogenicity. 2.MolecularIdentificationTechniques: a)PolymeraseChainReaction(PCR): PCR-basedtechniques,suchasrandomamplifiedpolymorphicDNA(RAPD)andamplifiedfragmentlengthpolymorphism(AFLP),wereemployedtoanalyzethegeneticdiversityoftheisolatedpathogens.Specificprimerstargetingconservedgenomicregionsofthepathogensweredesignedtoamplifyandidentifythetargetspecies.DNAsequencingwasperformedtovalidatetheidentificationresults. b)Real-timePCR(qPCR): qPCRisaquantitativeandrapidtechniquethatallowsforthedetectionandquantificationofthepathogenDNA.Specificprimersandprobestargetinguniquege
