以离子液体为流动相添加剂HPLC法测定钩藤碱和异钩藤碱的方法学研究 Title:MethodologicalInvestigationoftheDeterminationofUncarineAlkaloidsandIsouncarineAlkaloidsUsingHPLCwithIonicLiquidasMobilePhaseAdditive Abstract: Thedeterminationofuncarinealkaloidsandisouncarinealkaloidsisofgreatimportanceduetotheirpotentialpharmacologicalactivities.Thisresearchaimedtodevelopamethodologicalapproachusinghigh-performanceliquidchromatography(HPLC)withanionicliquid(IL)asamobilephaseadditive.Theeffectsofdifferentparameters,includingILconcentration,pH,mobilephasecomposition,andcolumntemperature,wereevaluatedtooptimizetheseparationanddetectionofthesealkaloids.ThedevelopedHPLCmethodshowedgoodsensitivity,linearity,precision,andaccuracyforthequantificationofuncarinealkaloidsandisouncarinealkaloids.Theproposedmethodcanbeappliedfortheanalysisofthesealkaloidsinvarioussamples,contributingtothequalitycontrolandpharmacologicalinvestigationofUncariaspecies. 1.Introduction: Uncaria,commonlyknownashooksvineorcat'sclaw,isagenusofplantswidelydistributedintropicalregions.Ithasbeenusedintraditionalmedicineforitspotentialanti-inflammatory,anticancer,andimmunomodulatoryactivities.Uncarinealkaloids,includinguncarineAandB,arethemajorbioactiveconstituentsfoundinUncariaspecies.Isouncarinealkaloids,suchasisouncarineEandF,areanalogsofuncarinealkaloidswithsimilarbiologicalactivities.Thedeterminationofthesealkaloidsischallengingduetotheirstructuralsimilarityandlowconcentrationsinnaturalmatrices.Therefore,areliableandsensitivemethodisneededfortheiranalysis. 2.Experimental: 2.1Chemicalsandreagents: Purestandardsofuncarinealkaloidsandisouncarinealkaloidswereobtainedfromreputablesuppliers.Theionicliquid(IL)wasselectedasthemobilephaseadditivetoenhancethechromatographicperformance.Othersolventsandreagents,includingmethanol,acetonitrile,andformicacid,wereofhighpurityandanalyticalgrade. 2.2Instrumentation: Ahigh-performanceliquidchromatography(HPLC)systemequippedwithaUV-Visdetectorwasemployedfortheanalysis.ThestationaryphaseusedwasaC18column.Themobilep
