RoleofProgranulininDiabeticNephropathyFootCellInjury: 1.Anti-inflammatoryEffects:Progranulinhasbeenshowntoattenuateinflammationthroughseveralmechanisms.Itsuppressestheproductionofpro-inflammatorycytokines,suchasinterleukin-6(IL-6)andtumornecrosisfactor-alpha(TNF-α),whichareknowntocontributetodiabeticnephropathy.Byinhibitingtheactivationofnuclearfactor-kappaB(NF-κB)signalingpathway,progranulineffectivelyreducestheexpressionofpro-inflammatorygenesinfootcells,thusprotectingagainstinjury. 2.RegulationofPodocyteFunction:Podocytesplayacrucialroleinmaintainingglomerularfiltrationbarrierintegrity.Diabeticnephropathyleadstopodocyteinjury,resultinginproteinuriaandfootcelldamage.Progranulinhasbeenshowntoenhancepodocytesurvivalandfunction.Itpromotespodocytemigrationandpreventspodocyteapoptosisbyactivatingthephosphoinositide3-kinase(PI3K)/Aktsignalingpathway.Additionally,progranulinstimulatestheexpressionofpodocyte-specificproteins,suchasnephrinandpodocin,whicharecriticalformaintainingpodocytestructureandfootcellintegrity. 3.PromotionofAngiogenesis:Diabeticnephropathyisassociatedwithimpairedangiogenesis,leadingtoreducedbloodflowtothefootanddelayedwoundhealing.Progranulincanstimulateangiogenesisthroughitsinteractionwithgrowthfactorsandextracellularmatrixcomponents.It
