CCDC106蛋白与p53蛋白相互作用的研究 Abstract: CCDC106isanovelproteinthatisfoundtointeractwiththetumorsuppressorp53,whichisinvolvedintheregulationofcellcycleprogressionandapoptosis.TheaimofthispaperistoreviewthecurrentliteratureontheinteractionbetweenCCDC106andp53,includingtheirexpressionpatterns,biologicalfunctionsandmechanismsofinteraction.Thepotentialimplicationsofthisinteractionincancerdevelopmentandtherapyarealsodiscussed. Introduction: p53isatumorsuppressorproteinthatplaysacriticalroleinregulatingcellcycleprogressionandpreventingthedevelopmentofcancer.Itisactivatedbyvariouscellularstresses,suchasDNAdamage,oxidativestressandoncogeneactivation,leadingtocellcyclearrest,apoptosisorsenescence.CCDC106isanewlyidentifiedproteinthatwasreportedtointeractwithp53inayeasttwo-hybridscreen.However,thebiologicalsignificanceofthisinteractionandtheunderlyingmechanismsremainunclear. ExpressionpatternsofCCDC106andp53: CCDC106isaproteinthatisubiquitouslyexpressedinvarioustissuesandcelllines,includingnormalandtumortissues.p53isalsowidelyexpressedindifferentcelltypes,withthehighestexpressioninthethymus,spleenandliver.Inaddition,p53expressionisoftenalteredincancercells,eitherthroughmutations,deletionsorpost-translationalmodifications,leadingtolossorgainoffunction. BiologicalfunctionsofCCDC106andp53: CCDC106isacytoplasmicproteinthatisinvolvedintheregulationofcellmigrationandinvasion.Ithasbeenshowntobindtotheactincytoskeletonandregulatefocaladhesionturnover,whichareimportantforcellmotility.Additionally,CCDC106hasbeenimplicatedintheregulationofautophagy,acellularprocessthatdegradesdamagedorunwantedproteinsandorganelles.p53isamultifunctionalproteinthatregulatesvariouscellularprocesses,includingcellcycleprogression,DNArepair,apoptosis,metabolismandsenescence.Itfunctionsasatranscriptionfactorthatactivatesorrepressestheexpressionofnumeroustargetgenesinvolvedintheseprocesses.
