Introduction: Extracellularpolysaccharides(EPS)arehigh-molecular-weightpolymersproducedbymanymicroorganisms,includinglacticacidbacteria.EPShavegainedattentionduetotheirdiversebiologicalactivities,includingantioxidantpropertiesthatcouldofferpotentialhealthbenefits.L.paracaseiVL8isknowntoproduceEPS,buttheoptimizationofEPSproductionconditionsanditsantioxidantpropertieshavenotbeenfullyexplored. Methods: 1.Strainandcultureconditions: L.paracaseiVL8wasobtainedfromaculturecollectionandmaintainedinMRSbroth.Thebacterialculturewasincubatedat37°Cwithcontinuousshakingfor24hours. 2.OptimizationofEPSproduction: TheproductionofEPSbyL.paracaseiVL8wasoptimizedforvariousparameters,includingcarbonsources,nitrogensources,pH,temperature,andincubationtime.TheEPSproductionwasquantifiedusingthephenol-sulfuricacidmethod. 3.Antioxidantactivityassay: TheantioxidantactivityoftheEPSwasdeterminedusingvariousinvitroassays,including1,1-diphenyl-2-picrylhydrazyl(DPPH)radicalscavengingassayandreducingpowerassay.Theresultswerecomparedwithstandardantioxidants,suchasascorbicacid. Results: 1.OptimizationofEPSproduction: TheoptimumconditionforEPSproductionbyL.paracaseiVL8wasdeterminedasfollows:glucoseasthecarbonsource,peptoneasthenitrogensource,pH6.5,temperature37°C,andincubationtimeof24hours.Undertheseoptimizedconditions,theyieldofEPSwassignificantlyincreased. 2.Antioxidantactivity: TheEPSproducedbyL.paracaseiVL8exhibitedsignificantantioxidantactivityinboththeDPPHradicalscavengingassayandreducingpowerassay.TheantioxidantactivityofEPSwascomparab
