PFOA对斑马鱼胚胎发育、行为和DNA损伤的毒性研究(英文) ToxicityofPFOAonDevelopment,Behavior,andDNADamageofZebrafishEmbryos Introduction Perfluorooctanoicacid(PFOA)isasyntheticorganofluorinecompoundthatiswidelyusedinseveralindustrialapplications.Duetoitspersistenceandbioaccumulationpotential,PFOAhasbecomeasubjectofconcernforitspotentialadverseeffectsonhumanhealthandtheenvironment.PreviousstudieshavereportedthetoxicityofPFOAonaquaticorganisms,includingzebrafish,whichiswidelyusedasamodelorganismfortoxicityassessment.Inthisstudy,weaimedtoinvestigatethetoxicityofPFOAonthedevelopment,behavior,andDNAdamageofzebrafishembryos. MaterialsandMethods ZebrafishembryoswereexposedtodifferentconcentrationsofPFOA(0,10,50,and100μg/L)from4hourspost-fertilization(hpf)to96hpf.Thedevelopmentofembryoswasassessedbymeasuringthehatchingrate,bodylength,andheartrate.Thebehaviorofzebrafishembryoswasdeterminedbymeasuringtheswimmingbehaviorusingavideotrackingsystem.TheDNAdamagewasevaluatedbymeasuringthelevelofDNAstrandbreaksusingthecometassay. Results OurresultsshowedthatexposuretoPFOAsignificantlydecreasedthehatchingrate,bodylength,andheartrateofzebrafishembryosinaconcentration-dependentmanner.TheswimmingbehaviorofzebrafishembryoswasalsoinfluencedbytheexposuretoPFOA,withasignificantincreaseinthedistancetraveledandvelocityatallconcentrationstested.Moreover,ourresultsshowedthatexposuretoPFOAinducedDNAdamageinzebrafishembryos,asevidencedbyasignificantincreaseinthelevelofDNAstrandbreaks. Discussion ThepresentstudyshowedthatPFOAhastoxiceffectsonthedevelopment,behavior,andDNAdamageofzebrafishembryos.Thedecreaseinhatchingrate,bodylength,andheartrateindicatesthatPFOAhasteratogeniceffectsonzebrafishembryos.Thechangesinswimmingbehaviorcouldberelatedtoalteredneurodevelopment,asthecentralnervoussystemisatargetofPFOAtoxicity.TheincreaseinDNAstrandbreakssuggeststhatPFOAhasgenotoxiceffectsonzebrafishembryos,whichcouldpotentiallyleadtomutationsandcarcinogenesis. Conclusion Inconclusion,PFOAhastoxiceffectsonthedevelopment,behavior,andDN
