蓝绿藻基因组DNA提取方法的比较研究(英文) AComparativeStudyontheDNAExtractionMethodsofCyanobacteriaGenomes Cyanobacteria,commonlyknownasblue-greenalgae,isadiversegroupofphotosyntheticorganismsthatarefoundinvariousaquaticandterrestrialenvironments.Theseorganismshaveattractedmuchattentionduetotheirabilitytofixatmosphericnitrogenandtheirpotentialasabiofuelsource.Understandingthegeneticmakeupoftheseorganismsiscrucialforunlockingtheirpotentialandstudyingtheirbiology.However,extractinghigh-qualityDNAfromcyanobacteriacanbechallengingduetothepresenceofcellwallstructuresandpigmentsthatinterferewithdownstreamapplications.Inthisstudy,wecompareandevaluatetheefficiencyofseveralDNAextractionmethodsforcyanobacteriagenomes. Methods WeselectedthreecommonlyusedDNAextractionmethodsforcyanobacteria:theCTAB(cetyltrimethylammoniumbromide)method,theQiagenDNeasyBloodandTissueKitmethod,andamodifiedSDS(sodiumdodecylsulfate)method.WeusedArthrospiraplatensisasthetestorganism,whichisafilamentouscyanobacteriumthatiswidelyusedforcommercialproductionofSpirulina.Thecellswereharvestedbycentrifugation,andtheDNAwasextractedaccordingtothemanufacturer'sinstructionsforeachmethod.WeevaluatedtheDNAqualityandquantitybyrunningagarosegelelectrophoresisandmeasuringtheabsorbanceat260nmand280nmusingaNanoDropspectrophotometer. Results TheCTABmethodyieldedhigh-qualityDNAwithahighpurityratio(A260/A280)of1.8-2.0.TheDNAwasintactandfreeofRNAcontamination,asshownbytheabsenceofRNAbandsinthegelelectrophoresis.Theyieldwasrelativelylow,withanaverageof55μg/gwetweightofcells.TheQiagenDNeasymethodresultedinaloweryieldofDNA(averageof38μg/gwetweightofcells)butwithhighpurityandgoodquality.TheDNAwasalsofreeofRNAcontamination.ThemodifiedSDSmethodyieldedthehighestamountofDNA(averageof69μg/gwetweightofcells),butwithalowerpurityratio(A260/A280)of1.6-1.7.RNAcontaminationwasobservedintheDNAsamples,asshownbythepresenceofRNAbandsinthegelelectrophoresis.
