近交系小鼠和中国小型猪血清和组织乳酸脱氢酶同工酶的电泳分析比较 摘要 近交系小鼠和中国小型猪都是经过多代近交选育而形成的纯合系。本研究旨在通过血清和组织乳酸脱氢酶(LDH)同工酶的电泳分析比较,探究近交对LDH同工酶的影响。结果显示近交系小鼠和中国小型猪的血清和组织LDH同工酶谱图有所差异,近交系小鼠呈现多个带位欠清晰,且缺少LDH-1、LDH-3和LDH-4带位,而中国小型猪则呈现较清晰的带位,且缺少LDH-4带位。结论为近交对LDH同工酶类的谱图会产生影响,但影响程度可能与物种和种类有关。 关键词:近交系小鼠;中国小型猪;血清;组织;乳酸脱氢酶;同工酶;电泳分析 Introduction Inbreeding,alsoknownasclose-breeding,isabreedingmethodinwhichahomozygousstrainisobtainedbyrepeatedinterbreedingoflinebredanimals.Multiplegenerationsofinbreedingoftenleadtotheaccumulationofhomozygousmutationsandthereductionandevenlossofgeneticdiversity,whichcanaffectthebiologicalcharacteristicsoforganisms,suchasfertility,growthrate,diseaseresistance,andimmunefunction.Theeffectofinbreedingonenzymeactivityhasbeenaresearchhotspot.Enzymeactivityisinfluencedbygeneticandenvironmentalfactors.ThephenotypevariationofLDHisoenzymesiscontrolledbygeneticfactorsandisresponsivetoenvironmentalfactors.LDHisanimportantenzymeforanaerobicglucosemetabolisminpyruvatemetabolism.Itsisoenzymedistributioncanreflectthedifferencesinenergymetabolismandtissuedifferentiationandhasbeenusedasabiologicalmarkerfordifferentcelltypesandtissues. MaterialsandMethods Experimentalanimals:TheexperimentalanimalsusedinthisstudywereinbredmiceandChineseminiaturepigs,respectively,whichwerebredbyinbreedingformorethan10generationsinthelaboratoryanimalcenter. Sampling:ThebloodsampleswerecollectedfromthetailveinsofmiceandtheneckveinsofChineseminiaturepigs.Theserumandtissuesamplesofliver,heart,andskeletalmusclewerecollectedandimmediatelystoredat-20°C. LDHisoenzymeanalysis:Theserumandtissuesampleswerehomogenizedinapotassiumphosphatesolutionandcentrifuged,andthesupernatantswereusedforelectrophoresis.LDHisoenzymeanalysiswasperformedusingpolyacrylamidegelelectrophoresis(PAGE)onaMini-PROTEANapparatus.
