ImmobilizationofGOxonsilkfibroinmembrane TheimmobilizationofGOxonsilkfibroinmembranecanbeachievedbyseveralmethods,includingphysicaladsorption,covalentbinding,andentrapment.Amongthesemethods,covalentbindingisthemostcommonlyusedmethodduetoitshighstabilityandefficientcatalyticactivity. ToimmobilizeGOxonsilkfibroinmembrane,firstly,thesilkfibroinmembranewasactivatedbytreatingitwithglutaraldehyde.Then,theactivatedsilkfibroinmembranewasincubatedwithGOx,andthecouplingreactionwascarriedoutundermildconditions.Finally,theimmobilizedGOxonsilkfibroinmembranewaswashedseveraltimestoremovetheunboundenzymemoleculesanddriedatroomtemperature. PropertiesofimmobilizedGOx TheimmobilizedGOxonsilkfibroinmembraneexhibitsseveralpropertiesthataredifferentfromthefreeGOxinsolution.Firstly,theimmobilizedGOxhasahigherthermalstability,whichisduetothehighstabilityofsilkfibroinmembraneandthestrongcovalentbondbetweenGOxandsilkfibroin.Secondly,theimmobilizedGOxhasahigherstoragestability,whichisbeneficialforlong-termstorageandpracticalapplications.Thirdly,theimmobilizedGOxexhibitsahigherresistancetopHanddenaturants,whichisimportantformaintainingitscatalyticactivityinharshconditions. ApplicationsofimmobilizedGOxinbiosensors TheimmobilizedGOxonsilkfibroinmembranehasbeenwidelyusedinbiosensorsforglucosedetection.Itcancatalyzetheoxidationofglucoseinthepresenceofoxygen,andthegeneratedhydrogenperoxidecanbedetectedelectrochemically.TheimmobilizedGOx-basedbiosensorshaveseveraladvantages,includinghighsensitivity,selectivity,andstability,andhavefoundapplicationsinclinicaldiagnosis,foodindustry,andenvironmentalmonitoring. Forexample,aGOx-basedbiosensorwasdevelopedforthedetectionofglucoseinhumanserumusingasilkfibroinmembraneasasupportingmaterial.Thebiosensor
