以水解液为原料的α-淀粉酶发酵工艺条件的研究 摘要 本文以水解液为原料,通过单因素实验和响应面实验优化了α-淀粉酶的发酵条件。结果表明,最佳条件为发酵温度35℃、发酵时间48h、初始pH值为6.5、α淀粉酶添加量为2.5g/L。在此条件下,α-淀粉酶的产酶量为690.45U/mL。最后,应用此条件进行了扩大实验,获得了较高的产量和活力。 关键词:α-淀粉酶;发酵条件;水解液;响应面实验 Introduction Alpha-amylasesareagroupofenzymeswhichcatalyzethehydrolysisofα-1,4-glycosidicbondsofstarch,glycogen,andrelatedpolysaccharides.Theyarewidelyusedinmanyindustries,suchasfood,fermentation,andtextile.Asaresult,itisimportanttooptimizeα-amylaseproductionprocesses.Inthisstudy,weaimedtooptimizethefermentationconditionsofα-amylaseusinghydrolysateastherawmaterial. Materialsandmethods α-amylaseproducingstrainBacillussubtiliswasusedinthisstudy.Hydrolysatewaspreparedbyhydrolyzingcornstarchwithhydrochloricacid.Singlefactorexperimentswerecarriedouttodeterminetheeffectofvariousfactorsonα-amylaseproduction.Thefactorsstudiedincludedtemperature,pH,fermentationtime,andα-amylaseconcentration.Acentralcompositedesignwasthenemployedtofurtheroptimizethefermentationprocess,andaresponsesurfacemethodologywasusedtoanalyzetheresults. Results Theresultsofsinglefactorexperimentsshowedthattheoptimaltemperatureforfermentationwas35℃,theoptimalpHwas6.5,theoptimalfermentationtimewas48hours,andtheoptimalα-amylaseconcentrationwas2.5g/L.Thecentralcompositedesignshowedthattheoptimalconditionswereatemperatureof35℃,pHof6.5,fermentationtimeof48hours,andα-amylaseconcentrationof2.5g/L.Undertheseconditions,theα-amylaseproductionwas690.45U/mL.Theseresultswereconfirmedintheenlargedexperiment,whichyieldedahighproductionandactivity.
