Introduction: Droughtstressisamajorlimitingfactoraffectingagriculturalproductivityworldwide,includingthegrowthanddevelopmentofbarleycrops.Barleyisknownforitsadaptabilitytovariousenvironmentalconditions,includingdroughtstress.Understandingthephysiologicalandmolecularresponsesofbarleytodroughtstressiscrucialfordevelopingdrought-tolerantvarieties.Inthisstudy,weaimtoexplorethephysiologicalchangesandwax-relatedgeneexpressionresponsesofdifferentbarleyvarietiesunderdroughtstressconditions. Methods: 1.Plantmaterial:Weselectedthreebarleyvarieties,A,B,andC,knownfortheirdifferentlevelsofdroughtresistance. 2.Droughtstresstreatment:Theselectedbarleyvarietiesweresubjectedtodroughtstressbywithholdingwatersupplyforaspecificperiod. 3.Physiologicalmeasurements:Wemeasuredvariousphysiologicalparametersincludingplantheight,leafarea,chlorophyllcontent,stomatalconductance,andwateruseefficiency. 4.RNAextractionandgeneexpressionanalysis:TotalRNAwasextractedfromtheleavesofdrought-stressedandcontrolplants.Theexpressionlevelsofwax-relatedgeneswereanalyzedusingquantitativereal-timePCR(qPCR).
