Introduction: CelluloseisthemostabundantorganiccompoundonEarthandiscomposedofglucoseunitslinkedbyβ-1,4-glycosidicbonds.Hydrolysisofcelluloserequirestheactivityofcellulases,whichareenzymescapableofbreakingdownthecomplexstructureofcelluloseintofermentablesugars.Bacillussiamensisisathermophilicbacteriumthathasthepotentialtoproducecellulaseenzymesefficiently.ThisstudyaimedtoscreenBacillussiamensisstrainsfortheircellulaseproductioncapabilityandoptimizetheconditionsforenhancedenzymeproduction. Methodology: 1.StrainScreening: -AcollectionofBacillussiamensisstrainswasobtainedfromaculturecollection. -Strainswereinitiallyscreenedforcellulaseproductiononcarboxymethylcellulose(CMC)agarplates. -PositivestrainswerefurtherevaluatedfortheircellulaseactivityusingtheDNS(3,5-dinitrosalicylicacid)method. 2.OptimizationofEnzymeProductionConditions: -Variouscarbonandnitrogensourceswerescreenedtodeterminethemostsuitableforcellulaseproduction. -Differentculturingconditions,includingpH,temperature,andagitationspeed,wereoptimizedtoenhanceenzymeproduction. -Theeffectofinducerssuchascelluloseandsophoroseoncellulaseproductionwasinvestigated. -EnzymeproductionwasmeasuredperiodicallyusingtheDNSmethod,andgrowthkineticsweremonitoredbymeasuringopticaldensity(OD600).
