Introduction: Parentageanalysisisarobusttoolfordeterminingthegeneticrelatednessbetweenindividualsandestablishingaccuratepedigrees.MicrosatellitesaretandemrepetitiveDNAsequencesthatareusedasgeneticmarkersinvariousorganismsduetotheirhighpolymorphismlevels.Thezebrafish(Daniorerio)isanidealmodelorganismforparentageanalysisduetoitsshortgenerationtime,highfecundity,andwell-characterizedgeneticmarkers. Methods: Forthisstudy,wecollectedtissuesamplesfrom100individualzebrafish,includingpotentialparentsandoffspring.DNAwasextracted,andPCRamplificationwasperformedusing10microsatelliteloci.Theamplifiedfragmentswereseparatedusingcapillaryelectrophoresisandanalyzedusingspecializedsoftwaretodeterminethenumberofrepeatsateachlocus.Thegenotypesofthepotentialparentsandoffspringwerecompared,andlikelihood-basedanalyseswereperformedtoassignparentage. Results: Theanalysisofthe10microsatellitemarkersresultedinaccurateparentageassignmentsforthezebrafishoffspring.Theaverageexclusionprobabilityforeachlocuswascalculated,indicatingthepowerofeachmarkerinexcludingfalselyassignedparents.Themeanexclusionprobabilityacrossalllociwasfoundtobe0.95,suggestingahighprobabilityofcorrectlyexcludingunrelatedindividualsaspotentialparents. Discussion: Microsatelliteshaveproventobeavaluabletoolinparentageanalysisinthezebrafish.The10microsatellitemarkersusedinthisstudyexhibitedsufficientpolymorphismandpowertoaccuratelydetermineparentage,providingarobustmethodforinvestigatinggeneticrelatednessandbreedingpatternsinzebrafi
