野苜蓿抑菌活性物质提取工艺的优化 Title:OptimizationofExtractionProcessforAntimicrobialActiveSubstancesfromWildAlfalfa Abstract: Wildalfalfa(Medicagosativa)isarichsourceofbioactivecompoundswithpotentialantimicrobialproperties.Inthisstudy,theextractionprocessforobtainingantimicrobialactivesubstancesfromwildalfalfawasoptimizedusingresponsesurfacemethodology(RSM).Theeffectsofextractionsolvents,extractiontime,solvent-to-solidratio,andextractiontemperatureontheantimicrobialactivityoftheextractwereinvestigated.Theresultsrevealedthattheoptimizedextractionprocessenhancedtheantimicrobialactivityofwildalfalfaextract,makingitapotentialcandidatefornaturalantimicrobialagents. 1.Introduction Theincreasingprevalenceofantibioticresistanceurgesthedevelopmentofalternativeantimicrobialagents.Naturalproducts,especiallythosederivedfromplants,havegainedattentionduetotheirdiversebioactivecompounds.Wildalfalfa,commonlyfoundinabundance,isknownforitstherapeuticproperties.Thisstudyaimstooptimizetheextractionprocessofantimicrobialactivesubstancesfromwildalfalfatoenhanceitsantimicrobialactivity. 2.MaterialsandMethods 2.1PlantMaterial Wildalfalfaplantswerecollectedfromruralareasandauthenticatedbyabotanist. 2.2ExtractionProcess Theextractionprocessinvolvedthefollowingsteps:(1)Preparationofwildalfalfasamplebydryingandgrindingitintoafinepowder.(2)Selectionofextractionsolvent.(3)OptimizationofextractionconditionsusingRSM.(4)Analysisofantimicrobialactivity. 2.3RSMOptimization Acentralcompositedesign(CCD)wasemployedtoevaluatetheeffectsofextractionparametersontheantimicrobialactivity.Aquadraticmodelwasusedtofittheexperimentaldataandoptimizetheextractionconditions. 3.ResultsandDiscussion 3.1SelectionofExtractionSolvent Differentsolvents(water,ethanol,methanol,andacetone)wereevaluatedfortheirefficiencyinextractingantimicrobialactivesubstancesfromwildalfalfa.Amongthem,ethanolshowedthehighestantimicrobialactivityandwasselectedastheextractionsolvent. 3.2OptimizationofExtractionConditions RSMallowedforthedeterminationoftheop
