一株产内切菊粉酶菌株培养条件的响应面优化 摘要:本研究以菊粉酶菌株为研究对象,采用响应面法进行优化,研究不同培养条件对菌株生长和内切菊粉酶产量的影响。结果表明,在温度、pH值和碳源浓度对生长和产酶影响的基础上,确定了最佳培养条件为:温度25℃,pH值6.5,碳源浓度0.5%。此项研究为菊粉酶菌株的高效生产提供了理论基础和实验指导。 关键词:菊粉酶菌株;响应面优化;温度;pH值;碳源浓度 Introduction: Inrecentyears,inulinasehasattractedattentionbecauseitcanhydrolyzeinulinintofructose,glucoseandothersubstances,whichcanbeusedintheproductionofsweeteners,biofuelsandotherfields.Inordertoobtainhigh-yieldinulinasestrains,itisimportanttooptimizethecultureconditions.Responsesurfacemethodology(RSM)isastatisticalmethodthatcanoptimizemultiplefactorsatthesametimeandiswidelyusedintheoptimizationofmicrobialcultureconditions. MaterialsandMethods: Theinulinase-producingstrainusedinthisstudywasisolatedfromsoil,anditsstrainnumberwasnamedINE216.Theculturemediumwascomprisedof:1%(w/v)inulin,0.2%(w/v)yeastextract,0.2%(w/v)peptone,0.1%(w/v)MgSO4·7H2O,0.05%(w/v)CaCl2·2H2Oanddistilledwater.Theoptimaltemperature,pHandcarbonsourceconcentrationwereselectedasthemainfactorsthatinfluencedthegrowthofthebacterialstrainandtheyieldofinulinase.AnexperimentwasdesignedusingRSM,theBox-Behnkendesign.Theexperimentalresultswereanalyzedbyregressionanalysisandthemeshdiagramwasdrawn. Results: TheBox-Behnkendesignmatrixforthethreefactorswasgenerated,andfifteenexperimentalrunswereperformedatrandom.Theresultsshowedthattheoptimaltemperature,pHandcarbonsourceconcentrationwere25℃,6.5and0.5%,respectively.Themaximuminulinaseactivitywas200U/mLattheseoptimalconditions.TheP-valueofthemodelwaslessthan0.05,indicatingthatallthemodeltermsweresignificant.Thecoefficientofdetermination(R2)was97.33%,indicatingthatthemodelwashighlyreliable. Conclusion: Inthisstudy,theresponsesurfacemethodologywasusedtooptimizethecultureconditionsofaninulinase-producingstrain.Theoptimaltemperature,pHandcarbonsourceconcentrationweredeterminedtobe25℃,6.5and0.5%,respectively.Thisstudyprovidestheoreticalfoundationsandexperimentalguidancefortheefficientproductionofinulinasestrains.
