鱼藤酮对大鼠肝毒性及其线粒体损伤机制的研究 Title:StudyonHepatotoxicityofFishVinegarKetoneanditsMechanismofMitochondrialDamageinRats Abstract: Therapiddevelopmentandwidespreadusageoftraditionalherbalmedicineshasraisedconcernsovertheirpotentialadverseeffectsonliverfunction.ThisstudyaimedtoinvestigatethehepatotoxicityofFishVinegarKetone(FVK)anditsunderlyingmechanismsofmitochondrialdamageinrats.Inthisstudy,maleWistarratswereorallyadministeredwithvaryingdosesofFVKfor28consecutivedays,andchangesinliverhistopathology,liverfunction,oxidativestressmarkers,andmitochondrialultrastructurewereevaluated.TheresultsrevealedthatFVKtreatmentcauseddose-dependentliverinjury,asevidencedbysignificantalterationsinliverenzymelevels,hepatichistopathologicalchanges,andcompromisedliverfunction.Furthermore,FVKinducedanimbalanceintheantioxidantsystem,leadingtoelevatedoxidativestress,asindicatedbysignificantlyincreasedlevelsofmalondialdehyde(MDA)anddecreasedlevelsofantioxidantenzymes.Notably,FVKtreatmentalsoresultedinmitochondrialdysfunction,includingdisruptionofmitochondrialmembranepotential,morphologicalabnormalities,anddeclinedmitochondrialrespiratoryfunction.Additionally,FVKexposurecausedasignificantdecreaseintheexpressionofmitochondrialfusionproteins(MFN1andMFN2),whileupregulatingtheexpressionofmitochondrialfissionprotein(DRP1).ThesefindingssuggestthatFishVinegarKetoneinduceshepatotoxicityinrats,possiblybyinducingoxidativestressandimpairingthemitochondrialdynamics.
