黑叶紫薇组培苗生根技术的研究(英文) ResearchonRootingTechnologyofBlack-LeafPlumBlossomTissueCultureSeedlings Abstract: Black-leafplumblossom(Prunuscerasifera)isapopularornamentalplantknownforitsdarkpurplefoliageandbeautifulpinkflowers.Tomeettheincreasingdemandforblack-leafplumblossomseedlings,tissueculturetechniqueshavebeendevelopedtopropagatetheplant.However,therootingofblack-leafplumblossomtissuecultureseedlingsremainsachallenge.Thisstudyaimstoinvestigatethefactorsaffectingtherootingofblack-leafplumblossomtissuecultureseedlingsandproposeoptimalrootingtechniques. 1.Introduction Black-leafplumblossomisasmalldeciduoustreeorshrubnativetoAsia,widelycultivatedforitsornamentalvalue.Itsdarkpurpleleavescontrastbeautifullywithitspinkflowers,makingitapopularchoiceforornamentalgardensandlandscapes.Tissuecultureoffersanefficientwaytomass-produceblack-leafplumblossomseedlingswithdesirabletraits,buttherootingprocessiscrucialforthesuccessfulestablishmentoftheseseedlingsinthefield. 2.MaterialsandMethods Inthisstudy,tissueculturetechniqueswereusedtopropagateblack-leafplumblossomseedlings.ExplantswereobtainedfromyoungshootsandculturedonMurashigeandSkoog(MS)mediumsupplementedwithvariousconcentrationsofplantgrowthregulators(PGRs).Differenttypesandconcentrationsofauxinsandcytokininsweretestedtoidentifytheoptimalrootingconditions.Therootingabilityoftheseedlingswasassessedbymeasuringrootlength,numberofroots,androotfreshweight. 3.Results Ourresultsshowedthatsuccessfulrootingofblack-leafplumblossomtissuecultureseedlingswasachievedbyusingacombinationofauxinandcytokinin.Indole-3-butyricacid(IBA)and6-benzylaminopurine(BA)werefoundtobethemosteffectivePGRsinpromotingrootformation.Amongdifferentconcentrationstested,0.5mg/LIBAand0.2mg/LBAyieldedthehighestrootlength,numberofroots,androotfreshweight.Theadditionofactivatedcharcoaltotherootingmediumfurtherenhancedrootingbyreducingtheoxidationofphenoliccompounds. 4.Discussion Theobservedrootingofblack-leafplumblossomtissuecultureseedlingsundercertainPGRcombinationscon
