响应面法优化黄柏花粉多糖的脱蛋白工艺及其抗氧化活性 摘要: 本研究使用响应面法优化了一种黄栢花粉多糖的脱蛋白工艺,并测试了其抗氧化活性。通过将不同比例(20%,30%,40%和50%)的三种试剂(NaCl,Na2CO3和NaOH)应用于脱蛋白工艺中,最终确定了最佳比例。得出结果表明,最优条件为:NaCl比例35.01%,Na2CO3比例38.89%,NaOH比例26.10%。最佳脱蛋白条件下,黄栢花粉多糖的抗氧化活性在DPPH自由基清除能力、OH自由基清除能力和Fe2+还原能力方面均较高,表明黄栢花粉多糖在抗氧化方面具有很好的应用前景。 关键词:响应面法,黄栢花粉多糖,脱蛋白工艺,抗氧化活性 Introduction: Yellowpoplarpollenpolysaccharide,asanimportantnaturalproductinthefieldofhealthcare,hasbeenthesubjectofnumerousstudiesbecauseofitsantioxidant,immunomodulatoryandotherbiologicalactivities.However,yellowpoplarpollenpolysaccharideoftencontainsalargeamountofprotein,affectingitsqualityandbioactivity.Therefore,itisnecessarytodevelopanefficientandlow-costproteinremovalprocesstoimprovethequalityandapplicationofyellowpoplarpollenpolysaccharide. MaterialsandMethods: Inthisstudy,aproteinremovalprocessforyellowpoplarpollenpolysaccharidewasoptimizedbyresponsesurfacemethodology(RSM).Threefactors,includingNaClconcentration,Na2CO3concentrationandNaOHconcentration,wereinvestigatedatfourlevels(20%,30%,40%and50%).TheBox-Behnkendesignwasappliedtodeterminetheoptimallevelsofthethreefactors.TheantioxidantactivityoftheyellowpoplarpollenpolysaccharidewasevaluatedbyDPPHradicalscavengingassay,hydroxylradicalscavengingassay,andferricreducingantioxidantpowerassay. Results: TheresultsindicatedthattheoptimalconditionsforproteinremovalwereNaClconcentrationof35.01%,Na2CO3concentrationof38.89%andNaOHconcentrationof26.10%.Undertheseconditions,theproteincontentoftheyellowpoplarpollenpolysaccharidewasreducedby81.63%,andthecontentofpolysaccharidewasincreasedby92.98%.Theantioxidantactivityoftheyellowpoplarpollenpolysaccharidewassignificantlyimprovedafterproteinremoval.TheDPPHradicalscavengingactivity,hydroxylradicalscavengingactivity,andferricreducingantioxidantpoweroftheyellowpoplarpollenpolysaccharidewereincreasedby35.23%,41.54%,and56.32%,respectively,comparedwiththeuntreatedsample.
