EGR1在RNA聚合酶Ⅰ指导的基因转录中的作用及机制研究 Title:TheRoleandMechanismofEGR1inRNAPolymeraseI-GuidedGeneTranscription Abstract: Genetranscriptionisahighlyregulatedandintricateprocessessentialforcellularfunctionanddevelopment.OnekeyplayerinthisprocessistheEarlyGrowthResponse1(EGR1)protein,whichhasbeenfoundtoplayacrucialroleinRNApolymeraseI(PolI)-guidedgenetranscription.Inthispaper,wereviewthecurrentunderstandingoftheroleandmechanismofEGR1inPolI-mediatedtranscription.Wediscussitsinvolvementingeneactivationandrepression,aswellasitsinteractionwithvarioustranscriptionalregulatorsandco-factors.Furthermore,weexploretheunderlyingmolecularmechanismsthatcontributetoEGR1'sfunctioninPolI-guidedtranscription,includingitsDNAbindingpropertiesandchromatinremodelingactivities.TheseinsightsprovideacomprehensiveunderstandingofEGR1'sroleingenetranscriptionandmayhaveimplicationsforfuturetherapeuticapproachestargetingthispathway. Introduction: RNApolymeraseI(PolI)isresponsiblefortranscriptionofribosomalRNA(rRNA)genes,whichareessentialforproteinsynthesisandcellgrowth.TranscriptionofrRNAisatightlyregulatedprocess,anddysregulationinPolI-guidedgenetranscriptionhasbeenlinkedtovariousdiseases,includingcancer.EGR1isaDNA-bindingtranscriptionfactorthatbelongstotheimmediateearlygenefamilyandhasbeenimplicatedinawiderangeofcellularprocesses,includingcellgrowth,differentiation,andapoptosis.Inrecentyears,increasingevidencehassuggestedthatEGR1playsanimportantroleinPolI-mediatedtranscription.Here,weexplorethecurrentliteratureontheroleandmechanismofEGR1inPolI-guidedgenetranscription. RoleofEGR1inPolI-GuidedTranscription: EGR1hasbeenshowntofunctionasbothatranscriptionalactivatorandrepressorinPolI-guidedtranscription.ItcanpositivelyregulatethetranscriptionofrRNAgenesbybindingtospecificDNAelementswithinthepromoterregionandrecruitingco-activatorstofacilitatePolIbindingandinitiation.Ontheotherhand,EGR1canalsorepressPolI-mediatedtranscriptionbyinteractingwithtranscriptionalrepressorsorcompetingwithactivatorsforDNAbindingsites.
