脓毒症大鼠血清Ang-2的动态变化及其与脓毒症肺损伤的关系 摘要: 脓毒症是一种常见的严重感染性疾病,其可引起各种器官系统的损伤。本研究旨在探究脓毒症大鼠血清Ang-2的动态变化以及其与脓毒症肺损伤的关系。通过建立脓毒症大鼠模型,检测不同时间点血清中的Ang-2水平,并观察大鼠肺组织的损伤情况。结果显示,脓毒症大鼠血清Ang-2水平在疾病早期逐渐上升,达到高峰后逐渐下降。同时,脓毒症大鼠肺组织显示不同程度的损伤,其中肺泡壁增厚和气体交换功能降低最为明显。进一步的分析表明,脓毒症大鼠血清Ang-2水平与肺损伤的严重程度呈正相关。因此,本研究认为,脓毒症大鼠血清Ang-2水平的动态变化可以用来评估脓毒症肺损伤的严重程度,有望成为脓毒症的重要诊断和治疗靶点。 关键词:脓毒症;Ang-2;肺损伤;动态变化;诊断;治疗 Introduction: Sepsisisacommonandseriousinfectiousdisease,whichcancausedamagetovariousorgansystems.Amongthecomplicationsassociatedwithsepsis,sepsis-inducedlunginjury(SILI)isaleadingcauseofmortalityincriticallyillpatients.AlthoughthepathophysiologyofSILIisnotfullyunderstood,previousstudieshavesuggestedthatangiogenesisplaysacrucialroleinthepathogenesisofthisdisorder.Specifically,theoverexpressionofangiopoietin-2(Ang-2),apotentangiogenicfactor,hasbeenlinkedtothedevelopmentandprogressionofSILI.However,thedynamicchangesinserumAng-2levelsandtheirrelationshipwithSILIhavenotbeenwellcharacterized. Methods: MaleSprague-Dawleyratswererandomlydividedintotwogroups:acontrolgroupandasepsisgroup.Thesepsisgroupwasinducedbycecalligationandpuncture(CLP).BloodsampleswerecollectedatdifferenttimepointsafterCLPsurgeryforthemeasurementofserumAng-2levels.Lungtissueswereharvestedforhistologicalanalysisandassessmentoflunginjury. Results: OurdatarevealedthatserumAng-2levelsincreasedgraduallyintheearlystageofsepsis,reachingapeaklevelat6hoursafterCLPsurgery.Afterwards,Ang-2levelsgraduallydecreasedduringthelatestageofsepsis.Incontrast,Ang-2levelsinthecontrolgroupremainedlowthroughouttheexperiment.Histologicalexaminationoflungtissuesshowedthatsepsis-inducedlunginjurywascharacterizedbythickeningofalveolarwalls,infiltrationofinflammatorycells,anddecreasedgasexchangefunction.Furthermore,theseverityofSILIwaspositivelycorrelatedwithserumAng-2levels. Conclusion: Inconclusion,ourresultssuggestthatserumAng-2levelsaredynamicallychangedduringsepsisandcanbeusedtoevaluatetheseverityofSILI.Therefore,targetingAng-2maybeapotentialtherapeuticstrategyforthetreat
